囊型包虫病所致过敏反应对白细胞介素‐17及抗体表达影响的实验研究 被引量:1

Experimental study on the effect of allergic reaction caused by cystic hydatid disease on interleukin⁃17 and antibody expression

摘要 目的探讨囊型包虫病所致过敏反应对IL‐17及抗体表达的影响。方法将120只雌性BALB/c小鼠按照随机数值大小分为3组:空白对照组(A组,45只)、囊型包虫感染模型组(B组,45只)、囊型包虫感染过敏反应组(C组,30只)。B组、C组建立包虫感染模型并观察3个月,模型建立成功后C组给予粗制囊液激发过敏反应,B组注射PBS。观察60 min后收集血清标本,使用流式细胞仪微珠阵列技术检测血清IL‐17水平。使用ELISA试剂盒检测小鼠血清免疫球蛋白E(immunoglobulin E,IgE)、免疫球蛋白G(immunoglobulin G,IgG)抗体水平。结果致敏前:B组IL‐17、IgE抗体及IgG抗体水平明显高于A组(P<0.05)。致敏后:B组IL‐17、IgE抗体及IgG抗体水平明显高于A组,C组IL‐17、IgE抗体及IgG抗体水平明显高于B组(P<0.05)。结论包虫过敏反应中,IL‐17作为炎症因子明显升高,IgE、IgG双抗体明显升高,提示IL‐17、IgE、IgG在过敏性反应发生、发展中起重要作用。 Objective To investigate the effect of allergic reaction caused by cystic hydatid disease on interleukin-17 (IL-17)and antibody expression. Methods One hundred and twenty female BALB/c mice were divided into three groups according to randomvalues: blank control group (group A, n=45), cystic hydatid infection model group (group B, n=45), and cystic hydatid infectionanaphylactic reaction group (group C, n=30). After the hydatid infection model was successfully established in groups B and C, group Cwas given crude capsular solution to stimulate anaphylactic reaction, and group B was injected with phosphate buffered saline (PBS).Serum samples were collected, and flow cytometry microbead array technology was used to detect the serum IL-17 levels in mice. Serumimmunoglobulin E (IgE) and immunoglobulin G (IgG) levels were detected using enzyme-linked immunosorbent assay (ELISA)kits. Results Before sensitization, the levels of IL-17, IgE and IgG antibodies in group B were significantly higher than those ingroup A (P<0.05). After sensitization, the levels of IL-17, IgE and IgG antibodies in group B were significantly higher than those ingroup A, and the levels of IL-17, IgE and IgG antibodies in group C were significantly higher than those in group B (P<0.05). Conclusions In hydatid anaphylactic reactions, IL-17 is significantly increased as an inflammatory factor, IgE and IgG antibodies are alsosignificantly increased, suggesting that IL-17, IgE and IgG play an important role in the occurrence and development of alnaphylacticreactions caused by cystic hydatidosis.
