摘要
对亮菌(Armillariella tabescens)多糖进行分级沉淀以研究亮菌多糖的单糖组成、分子量分布及其与活性之间的关系。采用不同浓度的乙醇分级沉淀,得到组分ATP-20、ATP-40、ATP-60、ATP-80。采用高效分子排阻色谱等方法进行多糖分子量和单糖组成分析,并对各组分进行抗氧化及免疫活性检测。结果表明,四个组分的重均分子量分别为8.959×105Da、1.277×106 Da、6.227×104 Da、1.778×104 Da;各组分均主要由葡萄糖、半乳糖、甘露糖、岩藻糖、木糖、阿拉伯糖、葡萄糖醛酸组成,但构成比例差别大。各组分均有明显的·OH自由基清除活性及促淋巴细胞增殖活性,其中ATP-80的羟自由基清除活性最高、促淋巴增殖活性最强;结合各个组分的得率及纯度,ATP-80更适合作为天然抗氧剂及免疫剂进行进一步的开发。
To investigate the monosaccharide composition,molecular weight distribution and their relationships to the activity of polysaccharides from Armillariella tabescens,fractional precipitation was carried out.The components ATP-20,ATP-40,ATP-60 and ATP-80 were obtained by fractional precipitation using ethanol at different concentrations.The weight-average molecular weight of the four components was 8.959×105,1.277×106,6.227×104,1.778×104,respectively.Analysis demonstrated that each component was mainly composed of glucose,galactose,mannose,fucose,xylose,arabinose and glucuronic acid,but the constituent ratio was greatly different.Each component showed obvious scavenging activities for hydroxyl radicals and lymphocyte proliferation-promoting activity.Among them,ATP-80 presented the highest scavenging activity for hydroxyl radicals and lymphocyte proliferation-promoting activity.Combined with the yield and purity of each component,ATP-80 is more suitable for the development of natural antioxidant and immune agent.
