注射用红花黄色素对大鼠股骨骨折愈合的治疗作用及机制研究 被引量:9

摘要 目的观察注射用红花黄色素对大鼠股骨骨折愈合的治疗作用,并分析相关的分子机制。方法建立大鼠股骨中段开放骨折模型,常规手术处理,实验组术后第1天开始给予注射用红花黄色素20 mg(溶于0.9%氯化钠注射液50 mL)缓慢静脉滴注,1次/d,共2周。对照组给予等量的0.9%氯化钠注射液缓慢静脉滴注。分别于术后第2、4、8周采用DR摄片观察骨折愈合情况。获取骨痂组织,提取RNA,逆转录,采用荧光定量聚合酶链式反应(polymerase chain reaction,PCR)及蛋白质印记观察骨形态发生蛋白2(bone morphogenetic protein 2,BMP-2)和骨形态发生蛋白7(bone morphogenetic protein 7,BMP-7)、RUNX家族转录因子2(RUNX family transcription factor 2,RUNX2)、MYC原癌基因(MYC proto-oncogene,MYC)及细胞周期蛋白D1(cyclin D1,CCND1)基因的表达。采用t检验或方差分析进行统计学分析。结果实验组大鼠第2、4、8周骨痂生成质量、速度及阴影密度均高于对照组,骨折愈合评分明显优于对照组(P<0.05)。实验组大鼠第2、4、8周骨痂组织中BMP-2、BMP-7、RUNX2、MYC及CCND1 mRNA及蛋白的表达量均高于同时间点对照组(P<0.05)。实验组大鼠第4、8周骨痂组织中BMP-2、BMP-7和RUNX2 mRNA及蛋白的表达量均高于第2周,且第4周的表达量达到巅峰(P<0.05)。实验组大鼠第2、4、8周骨痂组织中MYC和CCND1 mRNA及蛋白的表达量呈上升趋势,且第8周的表达量达到巅峰(P<0.05)。结论注射用红花黄色素促进大鼠股骨骨折愈合,其作用机制可能与上调骨痂组织中BMP-2、BMP-7、RUNX2、MYC及CCND1基因表达相关。 Objective To observe the therapeutic effect of safflor yellow injection on the healing of femoral fracture in rats,and analyze related molecular mechanism.Methods A open femoral fracture model of rat was established and routinely treated.The experimental group was treated with 20 mg of safflor yellow injection(dissolved in 50 mL of0.9%sodium chloride)for slow intravenous drip on the first day after surgery at 1 time/d,a total of 2 weeks.The control group was given an equal amount of 0.9%sodium chloride.DR imaging was used to observe the fracture healing at 2nd,4th and 8th weeks after surgery.The epiphyseal tissues were collected,RNA was extracted,and reverse transcription was performed.The expression of bone morphogenetic protein 2(BMP-2)and bone morphogenetic protein 7(BMP-7),RUNX family transcription factor 2(RUNX2),MYC proto-oncogene(MYC)and cyclin D1(CCND1)genes were observed by quantitative real-time PCR and Western blotting.Statistical analysis was assessed by using Student’s t test or analysis of variance.Results The rats in the experimental group had higher quality,speed,and shadow density of epiphysis than that in the control group at 2nd,4th and 8th weeks,and the fracture healing scores were significantly higher in the experimental group than the control group(P<0.05).The mRNA and protein expression levels of BMP-2,BMP-7,RUNX2,MYC and CCND1 in the piphyseal tissues of the experimental group were higher than those in the control group at 2nd,4th and 8th weeks(P<0.05).The mRNA and protein expression levels of BMP-2,BMP-7 and RUNX2 in the epiphyseal tissues of rats of the experimental group at 4th and 8th weeks were higher than those in the 2nd week,and the expression levels reached the peak in the 4th week(P<0.05).The mRNA and protein expression levels of MYC and CCND1 in the epiphyseal tissues of the experimental group showed an upward trend in the 2nd,4th and 8th weeks,and the expression levels reached the peak in the 8th week(P<0.05).Conclusion Safflor yellow injection promotes the healing of femoral
