鸡冠花药材HPLC指纹图谱及黄酮类成分含量测定研究 被引量:2

摘要 目的:建立鸡冠花药材HPLC指纹图谱测定方法,对市售鸡冠花药材的质量进行较全面的评价。方法:采用Waters HSS T3色谱柱(4.6 mm×250 mm,5μm),以乙腈-0.2%磷酸溶液(含0.2%三乙胺)为流动相进行梯度洗脱,指纹图谱检测波长为290 nm,山柰素、异鼠李素、山奈酚-4’-O-甲醚含量测定检测波长为365 nm,柱温为30℃,流速为1.0 mL/min。采用相似度评价、聚类分析对15批鸡冠花药材指纹图谱进行研究。结果:建立了鸡冠花药材的HPLC指纹图谱,确定了8个共有峰;通过相似度评价、聚类分析结果可知15批鸡冠花药材质量差异不明显,并比较了样品中山柰素、异鼠李素、山奈酚-4’-O-甲醚的含量。结论:该方法为鸡冠花药材的质量控制提供了较为全面、有效的快速评价方法。 Objective: To establish a method for the determination of HPLC fingerprint of Jiguanhua(celosiae cristatae flos), and to evaluate the quality of commercial Jiguanhua(celosiae cristatae flos). Methods: A Waters HSS T3 column(4.6 mm×250 mm, 5 μm) was used with acetonitrile-0.2% phosphoric acid solution(containing 0.2% triethylamine) as the mobile phase for gradient elution. The detection wavelength of fingerprint was 290 nm, and the detection wavelength of kaempferol, isorhamnetin and kaempferol-4 ’-o-methyl ether was 365 nm.The column temperature was 30 ℃, and the flow rate was 1.0 mL/min. The fingerprints of 15 batches of Jiguanhua(celosiae cristatae flos) were studied by similarity evaluation and cluster analysis. Results: The HPLC fingerprint of Jiguanhua(celosiae cristatae flos) was established, and 8 common peaks were determined. The results of similarity evaluation and cluster analysis showed that the quality of 15 batches of Jiguanhua(celosiae cristatae flos) was not significantly different, and the contents of kaempferin, isorhamnetin and kaempferol-4’-O-methyl ether in the samples were compared. Conclusion: This method provides a more comprehensive and effective rapid evaluation method for the quality control of Jiguanhua(celosiae cristatae flos).
