不同产地大蓟、小蓟的指纹图谱研究 被引量:4

Study on the Fingerprint of Daji (Cirsii Japonici Herba seu Radix) and Xiaoji (Cirsii Herba) from Different Habitats

摘要 目的:分别建立不同产地大蓟、小蓟HPLC指纹图谱,并比较其异同,为其质量控制提供参考依据。方法:色谱柱Diamonsil C18柱(4.6 mm×250 mm,5μm),以乙腈-0.1%磷酸水溶液为流动相进行梯度洗脱;流速为1 mL/min;检测波长为330 nm;柱温为30℃;进样量为10μL。采用相似度评价方法对指纹图谱进行研究。结果:10批大蓟共有18个共有峰,并指认其中6种,分别为新绿原酸、绿原酸、隐绿原酸、咖啡酸、蒙花苷和柳穿鱼叶苷;15批小蓟共有16个共有峰,并指认其中6种,分别为新绿原酸、绿原酸、隐绿原酸、咖啡酸、芦丁和蒙花苷。不同产地的大蓟、小蓟药材具有稳定且特征的指纹图谱。结论:该方法稳定、可靠,可为大蓟、小蓟的质量控制提供一定的依据。 Objective: To establish the fingerprints of Daji(cirsii japonici herba seu radix) and Xiaoji(cirsii herba) from different habitats by HPLC, and to compare their similarities and differences, so as to provide reference for quality control of Daji(cirsii japonici herba seu radix) and Xiaoji(cirsii herba). Methods: The chromatographic column Diamonsil C18(4.6 mm×250 mm, 5 μm) was used for gradient elution with acetonitrile-0.1%phosphoric acid solution as mobile phase at a flow rate of 1 mL/min. The detection wavelength was 330 nm, the column temperature was 30 ℃, and the injection volume was 10 μL. The similarity evaluation method was used to study the fingerprint. Results: There were 18 common peaks in 10 batches of Daji(cirsii japonici herba seu radix), and 6 of them were identified as neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, caffeic acid, linarin and andrographolide. There were 16 common peaks in 15 batches of Xiaoji(cirsii herba), and 6 of them were identified as neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, caffeic acid, rutin and linarin. There are stable and characteristic fingerprints of Daji(cirsii japonici herba seu radix) and Xiaoji(cirsii herba) from different habitats. Conclusion: The method is stable and reliable, and can provide a certain basis for quality control of Daji(cirsii japonici herba seu radix) and Xiaoji(cirsii herba).
