摘要
目的:研究茵杞调脂饮改善非酒精性脂肪肝病(NAFLD)大鼠脂质代谢的作用及机制。方法:48只雄性SD大鼠随机分为正常组,模型组,茵杞调脂饮低、中、高剂量组及辛伐他汀对照组,各8只。正常组喂养普通饲料,其余组喂养高脂饲料,12周后模型组灌胃0.9%氯化钠溶液,各药物组灌胃相应剂量药物,8周后检测血清ALT、AST、TC、TG及肝组织TC、TG、LP、TNF-α含量,HE、油红O染色检测肝组织脂肪变性程度,RT-PCR检测肝组织LXRα、FAS mRNA表达。结果:与正常组比较,模型组大鼠血清ALT、AST、TC、TG及肝组织TC、TG、LP、TNF-α,LXRα、FAS mRNA显著升高(P<0.01),肝组织出现显著的细胞脂肪变性及空泡样变;与模型组比较,各药物组大鼠血清ALT、AST、TC、TG及肝组织TC、TG、LP、TNF-α,LXRα、FAS mRNA显著降低(P<0.05,P<0.01),肝细胞脂肪变性及空泡样变得到明显改善;与辛伐他汀组比较,茵杞调脂饮高剂量组血清ALT、AST,肝组织LP、TNF-α及LXRα、FAS mRNA显著降低(P<0.05)。结论:茵杞调脂饮可以调节NAFLD大鼠的脂代谢,减少脂质沉积,减轻氧化应激,其机制可能与抑制LXRα、FAS的表达有关。
Objective:To investigate the effects of Yinqi Tiaozhi Decoction on improving lipid metabolism in rats with non-alcoholic fatty liver disease(NAFLD).Methods:Forty-eight healthy male SD rats were randomly divided into normal group,model group,low-dose,middle-dose and high-dose Yinqi Tiaozhi Decoction group and Simvastatin positive drug control group,8 rats each group.The normal group received standard diet and the rest groups were fed with high-fat diet.After 12 weeks,the model group was given normal saline,and drug groups was given the corresponding dose of drugs.After 8 weeks,all of the rats were sacrificed.The observing items including ALT,AST,TC and TG levels in serum;TC,TG,TNF-α,LP content in liver tissue;HE staining and oil red O staining were used to detecting the degree of hepatic steatosis;RT-PCR testing LXRαand FAS mRNA expression in liver.Results:Compared with normal group,the levels of ALT,AST,TC,TG in serum,TC,TG,LP,TNF-αand LXRα,FAS mRNA in liver in the model group were remarkably elevated(P<0.01);liver tissue showed significant hepatocyte steatosis and vacuolar degeneration.Compared with the model group,drug groups of levels of ALT,AST,TC,TG in serum,TC,TG,LP,TNF-αand LXRα,FAS mRNA in liver were decreased(P<0.05,P<0.01);hepatocyte steatosis and vacuolar degeneration improved significantly.Compared with the simvastatin group,the high-dose group of Yinqi Tiaozhi Decoction showed a significant efficacy in serum ALT,AST,and liver tissue LP,TNF-α,LXRα,FAS mRNA decreased significantly(P<0.05).Conclusion:Yinqi Tiaozhi Decoction can regulate lipid metabolism,reduce lipid deposition and oxidative stress in rats with nonalcoholic fatty liver disease.The mechanism may be related to the inhibition of LXRαand FAS expression.
