摘要
目的观察白藜芦醇对白细胞介素(IL)-1β诱导的椎间盘髓核细胞增殖及炎性反应的影响。方法通过酶序贯消化法获得大鼠椎间盘髓核细胞,并将细胞分为正常组、IL-1β组(20μg/L)、白藜芦醇低、中、高剂量组(IL-1β+12.5、25.0、50.0μmol/L白藜芦醇)。细胞计数试剂盒(CCK-8)实验检测细胞活力、流式细胞术检测细胞凋亡率,实时定量聚合酶链反应(Real-time PCR)检测肿瘤坏死因子-α(TNF-α)、IL-6 mRNA表达,蛋白质印迹法(Western blot)检测核因子-κB(NF-κB)蛋白表达量,组间比较采用LSD-t检验。结果白藜芦醇低、中、高剂量组与IL-1β组比较,细胞活力(0.396±0.040、0.435±0.043、0.517±0.052比0.362±0.036,t=5.856、6.437、6.893,P均<0.05)显著提高;细胞早期[(15.23±1.53)%、(9.54±1.05)%、(3.56±0.36)%比(18.63±1.86)%,t=5.587、6.394、6.885,P均<0.05]及晚期凋亡率[(14.29±1.43)%、(8.34±0.83)%、(4.39±0.44)%比(19.56±1.96)%,t=5.136、6.483、6.679,P均<0.05],TNF-α(3.63±0.36、2.89±0.29、1.96±0.20比4.56±0.36,t=5.436、6.135、5.378,P均<0.05),IL-6(2.98±0.29、2.16±0.22、1.86±0.19比3.78±0.38,t=5.842、5.689、6.234,P均<0.05)mRNA表达量及p-NF-κB p65(0.29±0.03、0.169±0.01、0.065±0.01比0.66±0.06,t=5.632、6.137、6.841,P均<0.05)蛋白表达量均显著降低。结论白藜芦醇通过降低NF-κB活性抑制IL-1β诱导的椎间盘髓核细胞增殖及炎性反应。
Objective To explore effect of resveratrol on proliferation and inflammatory response of nucleus pulposus cells of intervertebral disc induced by interleukin(IL)-1β.Methods The cells were divided into normal group,IL-1βgroup(20μg/L),low,medium and high dose resveratrol group(IL-1β+12.5,25.0,50.0μm resveratrol).Cell viability was detected by CCK-8 methods.Cell apoptotic rate was detected by flowcytes methods.The content of tumor necrosis factor alpha(TNF-α)and IL-6 was detected by Realtime PCR.The expression of nuclear factor-κB(NF-κB)p65 was detected by Western blotting.Results Compared with IL-1βgroup,Cell viability was increased in low,medium and high dose resveratrol group(0.396±0.040,0.435±0.043,0.517±0.052 vs.0.362±0.036,t=5.856,6.437,6.893,P all<0.05).Cell early apoptotic rate[(15.23±1.53)%、(9.54±1.05)%、(3.56±0.36)%vs.(18.63±1.86)%,t=5.587,6.394,6.885,P all<0.05)]and late apoptotic rate[(14.29±1.43)%、(8.34±0.83)%、(4.39±0.44)%vs.(19.56±1.96)%,t=5.136,6.483,6.679,P all<0.05)],and the expression of TNF-α(3.63±0.36,2.89±0.29,1.96±0.20 vs.4.56±0.36,t=5.436,6.135,5.378,P all<0.05),IL-6(2.98±0.29,2.16±0.22,1.86±0.19 vs.3.78±0.38,t=5.842,5.689,6.234,P all<0.05)mRNA,the expression of p-NF-κB p65(0.29±0.03,0.169±0.01,0.065±0.01 vs.0.66±0.06,t=5.632,6.137,6.841,P all<0.05)was decreased in low,medium and high dose resveratrol group.Conclusion Resveratrol could inhibit the proliferation and inflammatory response of nucleus pulposus cells induced by IL-1βby reducing NF-κB activity.
