补肾活血复方对心肌梗死后心力衰竭大鼠CaMKⅡ的影响研究 被引量:3

摘要 目的:探究补肾活血复方对心梗后心衰大鼠钙/钙调蛋白依赖的蛋白激酶Ⅱ(CaMKⅡ)的影响。方法:选取12周龄雄性Wistar大鼠80只,随机取20只为假手术组仅挂线,不结扎。另60只大鼠结扎左冠状动脉前降支,术后予喂食减半配合游泳,造模成功后随机分为模型组、中药组和西药组。西药组给予卡托普利,中药组给予补肾活血复方,正常组和模型组均给予同等剂量蒸馏水。用药4周后,超声检测后计算左心室舒张末期内径(LVEDD)、收缩末期左心室内径(LVESD)、左心室射血分数(EF)和左心室缩短分数(FS),酶联免疫法测定大鼠血清脑利钠肽(BNP)表达水平,免疫组化法检测大鼠心肌组织中CaMKⅡ表达水平。结果:用药4周后,与假手术组相比,模型组LVEDD、LVESD升高,EF、FS降低,血清BNP明显增高,心肌CaMKⅡ表达增高,差异有统计学意义(P<0.05);和模型组相较,中药组和西药组大鼠LVEDD、LVESD下降,EF、FS增高,中药组、西药组血清BNP明显降低,心肌CaMKⅡ表达降低,差异有统计学意义(P<0.05)。结论:补肾活血复方对心肌中CaMKⅡ的表达有明显抑制作用,进一步延缓心室重构,改善慢性心衰。 Objective:To explore Bushen Huoxue decoction heart failure after myocardial infarction rats calcium/calmodulin dependent protein kinaseⅡ(CaMKⅡ).Methods:8012-week-old male Wistar rats were selected,and 20 were randomly selected into the sham operation group,in which only the wires were attached without ligation.The remaining 60 were ligated with the anterior descending branch of the left coronary artery,followed by diet reduction and exhaustion swimming.After remodeling sucessfully,they will be randomly separated into model group,traditional Chinese medicine group and western medicine group.Western medicine group was given captopril,Chinese medicine group was given Bushen Huoxue decoction,normal group and model group were given the same dose of distilled water.Ultrasonic testing rats after 4 weeks,left ventricular end-diastolic diameter(LVEDD),contract at the end of the left ventricular diameter(LVESD)and calculate the left ventricular ejection fraction(EF)and left ventricular short axis shortening fraction(FS),enzyme-linked immune method of rat serum brain natriuretic peptide(BNP)expression level,immunohistochemical method to detect myocardial tissue of rats in CaMKⅡexpression level.Results:After 4 weeks,compared with the control group,model group increased LVEDD,LVESD,EF,FS were reduced,increased serum BNP,myocardial CaMKⅡexpression rise,statistical calculations showed that it makes sense(P<0.05).By contrast model group,traditional Chinese medicine and western medicine group rats reduced LVEDD,LVESD,EF,FS,traditional Chinese medicine,western medicine group obviously decreased serum BNP,myocardial CaMKⅡexpression were reduced,statistical calculations showed that it made sense(P<0.05).Conclusion:Bushen Huoxue decoction in myocardial CaMKⅡexpression has obvious inhibitory effect,further delay ventricular remodeling and improve chronic heart failure.
