摘要
目的探讨依普利酮对慢性心力衰竭大鼠血流动力学和运动能力的影响。方法将18只成年健康清洁级SD雄性大鼠随机分为空白组、心力衰竭组与依普利酮组,每组6只大鼠。心力衰竭组与依普利酮组均采用结扎左冠状动脉前降支建立心力衰竭模型,依普利酮组在心力衰竭模型建立后2周给予依普利酮20 mg/(kg·d)灌胃,连续应用4周。记录大鼠不同时间点的血流动力学和运动能力。结果所有大鼠都成活,心力衰竭组与依普利酮组实验2周、6周的收缩压与舒张压均明显高于空白组(P<0.05),依普利酮组实验6周的收缩压与舒张压均明显低于心力衰竭组(P<0.05)。实验6周,心力衰竭组与依普利酮组左室收缩末期内径(LVESD)明显高于空白组(P<0.05),依普利酮组LVESD低于心力衰竭组(P<0.05)。实验6周心力衰竭组与依普利酮组左室内压最大上升速率(+dp/dtmax)和左室内压最大下降速率(-dp/dtmax)值均低于空白组(P<0.05),依普利酮组+dp/dtmax、-dp/dtmax均高于心力衰竭组(P<0.05)。结论采用结扎左冠状动脉前降支建立心力衰竭模型能反映大鼠的心力衰竭状况,依普利酮可改善慢性心力衰竭大鼠血流动力学状况,提高心功能与运动能力。
Objective To investigate the effects of eplerenone on hemodynamics and exercise capacity in rats with chronic heart failure.Methods Eighteen adult healthy and clean SD male rats were divided into three groups:blank group,heart failure group,and eplerenone group,with 6 rats in each groups.The heart failure model in heart failure group and eplerenone group were established in by ligation of the left anterior descending coronary artery.The rats in eplerenone group were given eplerenone 20 mg/(kg·d)after the heart failure model establishment of 2 weeks,and the treatment were lasted for 4 weeks.The hemodynamics and exercise capacity of the rats at different time points were recorded.Results All rats were survived,the systolic and diastolic blood pressures in heart failure group and eplerenone group were significantly higher than those in blank group at 2 and 6 weeks(P<0.05).The systolic and diastolic pressures in eplerenone group were significantly lower than those of the heart failure group at 6 weeks(P<0.05).The left ventricular end systolic diameter(LVESD)of heart failure group and eplerenone group were significantly higher than that of blank group at 6 weeks(P<0.05),and the LVESD of eplerenone group was lower than that of the heart failure group(P<0.05).The+dp/dtmax and-dp/dtmax values of heart failure group and eplerenone group were lower than those of blank group at 6 weeks(P<0.05),and the+dp/dtmax and-dp/dtmax values of eplerenone group were higher than those of heart failure group(P<0.05).Conclusion The establishment of heart failure model by ligating the left anterior descending coronary artery can reflect the heart failure condition of rats.The application of eplerenone in chronic heart failure rats can improve hemodynamics and improve cardiac function and exercise capacity.
