摘要
为博落回饮片的质量控制及标准建立提供参考,以博落回为研究对象,采用薄层色谱法和高效液相色谱法,建立博落回药材的薄层鉴别方法和血根碱含量的测定方法,参照2015年版《中国药典》(4部)要求,测定博落回饮片的水分、灰分、浸出物含量,初步拟定博落回饮片质量控制标准。结果表明:薄层色谱斑点清晰,分离度好;血根碱的含量在0.27%~0.71%,平均0.43%,在0.08~1.04μg线性范围内与峰面积呈良好线性关系(r=0.9997);平均回收率为102.24%,该方法简便,专属性强,重复性好,可用于博落回饮片的质量检测;初步拟定博落回饮片标准为水分含量≤13.0%、总灰分≤8.0%、水溶性浸出物含量≥17.0%、醇溶性浸出物含量≥17.0%、血根碱含量≥0.20%。
To provide a reference for the quality control and standard establishment of Macleaya cordata decoction pieces,TLC and high performance liquid chromatography were used to establish the thin-layer identification method of M.cordata medicinal materials and the content of sanguinarine.The moisture,ash,and extract content of M.cordata decoction pieces were measured by referring to the requirements of Part 4 of the Chinese Pharmacopoeia(2015 eds),and preliminarily draft the quality control standards for M.cordata decoction pieces.Results:The TLC spots were clear and the resolution was good;The content of sanguinarine ranged from 0.27%to 0.71%,with an average of 0.43%,and there was a good linear relationship with the peak area in the linear range of 0.08 to 1.04μg(r=0.9997);The average recovery rate was 102.24%.The method was simple,specific,and reproducible.It could be used for the quality detection of M.cordata decoction pieces.The preliminary draft standards for M.cordata decoction pieces included moisture content≤13.0%,total ash content≤8.0%,water-soluble extract content≥17.0%,alcohol-soluble extract content≥17.0%and sanguinarine content≥0.20%.
