摘要
骨关节炎 (osteoarthritis,OA) 是一种以关节软骨退行性变、软骨下骨骨质增生为特征的慢性、退行性关节疾病,而软骨细胞外基质代谢紊乱是关节软骨病理改变的关键环节[1]。大量的研究发现,组蛋白乙酰化酶 (histone deacetylases,HDACs) 直接调控软骨细胞组蛋白乙酰化水平,进而影响细胞活性以及调控软骨细胞外基质表达,从而在骨关节发生、发展中扮演着重要的角色[2]。
Osteoarthritis (OA) is a joint disease characterized by articular cartilage degeneration and subchondral bone sclerosis.Its occurrence is related to obesity,age,inflammation,trauma,genetics and other factors.The main pathological changes of OA are chondrocyte apoptosis and cartilage extracellular matrix degradation.Histone acetylation modification is an important process in epigenetic regulation,which can regulate gene expressions at the transcriptional level and thus plays an important role in a variety of diseases’ progression.In recent years,many studies have confirmed that histone acetylation functions in maintaining the homeostasis of cartilage,and its abnormal regulation can lead to the development of osteoarthritis.Histone deacetylases (HDACs) are important enzymes for histone deacetylation modification,and they have gradually become important target molecules to study cartilage degeneration.Moreover,many literatures have reported that HDACs-related inhibitors (histone deacetylase inhibitors,HDACi) have the potential to alleviate the destruction of articular cartilage and inhibit the progression of OA.In addition,many clinical studies have been carried out to explore the therapeutic roles of HDACi in OA.This review aims to summarize the biological characteristics and molecular mechanisms of HDACs in articular cartilage degeneration,and the effects of HDACi on OA treatment,thereby providing new ideas for the prevention and treatment of OA.
