调治天癸方对少弱精子症模型大鼠Bax、Bcl-2表达的影响 被引量:3

Effect of Tiangui Regulating Prescription on Expression of Bax and Bcl-2 in Model Rat of Oligoasthenozoospermia

摘要 目的:观察调治天癸方对少弱精子症大鼠睾丸生精细胞Bcl-2/Bax蛋白表达的影响。方法:60只SD大鼠随机选取10只作为空白组,余50只大鼠均采用环磷酰胺35 mg·kg^-1·d^-1腹腔注射法复制少弱精子症模型,连续注射5 d。造模后将大鼠随机分为模型组,生精胶囊组,调治天癸方高剂量组、中剂量组、低剂量组,每组10只,空白组同时腹腔注射等体积生理盐水。造模第6天开始,各组以每100 g体质量灌胃相应药物,生精胶囊组以0.0432 g灌胃,调治天癸方高剂量组(5.076 g)、中剂量组(2.538 g)、低剂量组(1.269 g)分别以调治天癸方灌胃,空白组及模型组给予等体积生理盐水灌胃,各组每天均按时灌胃1次,连续2周。实验结束后将大鼠全部脱颈处死,经双侧附睾、输精管中采集大鼠精子标本进行精液常规分析;摘取睾丸,采用免疫组织化学法检测睾丸组织Bcl-2、Bax蛋白的表达。结果:模型组精子质量下降最明显,睾丸组织中Bax蛋白显著升高,Bcl-2蛋白显著降低;与模型组比较,各治疗组精子数量、a+b级精子比例、精子活动率均显著提高,睾丸组织Bax蛋白表达水平显著降低,Bcl-2蛋白表达水平显著增高;且调治天癸方疗效优于生精胶囊,差异均有统计学意义(P<0.05)。结论:调治天癸方可通过调控睾丸组织Bcl-2及Bax蛋白的表达,减少生精细胞凋亡,从而改善少弱精子症大鼠的精液质量。 Objective:To observe the effect of Tiaozhi Regulating Prescription on the expression of Bcl-2/Bax protein in spermatogenic cells of oligoasthenozoospermia rats.Method:60 SD rats were randomly selected as the blank group,and the remaining 50 rats were injected with cyclophosphamide 35 mg·kg^-1·d^-1 intraperitoneally to replicate the oligoasthenospermia model.The injection was continued for 5 days and the model was created.The rats were randomly divided into model group,Shengjing Capsule group,Tiangui Regulating Prescription high-dose group,middle-dose group,and low-dose group,with 10 rats in each group.The blank group was injected intraperitoneally with an equal volume of normal saline.Beginning on the 6th day of modeling,each group was given the corresponding drugs per 100 g body weight,the Shengjing Capsule group was given 0.0432 g,and the Tiangui Regulating Prescription high-dose group(5.076g),medium-dose group(2.538 g),and low-dose Groups(1.269 g)were given Tiaozhi Regulating Prescription by gavage.The blank group and model group were gavage with equal volume of normal saline.Each group was gavage once a day on time for 2 weeks.After the experiment,all the rats were sacrificed by neck removal.Sperm samples were collected from the bilateral epididymis and vas deferens for routine semen analysis;the testes were taken,and the expression of Bcl-2 and Bax protein in testicular tissue was detected by immunohistochemistry.Results:The sperm quality of the model group decreased the most,the Bax protein in the testis tissue was significantly increased,and the Bcl-2 protein was significantly decreased.Compared with the model group,the sperm count,the ratio of a+b grade sperm,and the sperm motility rate of each treatment group were significantly increased The expression level of Bax protein in testis tissue was significantly reduced,and the expression level of Bcl-2 protein was significantly increased.Tiangui Regulating Prescription was better than Shengjing Capsules,and the difference was statistically significant(P<0.
