摘要
目的探讨FOXP3^+细胞在胃癌不同部位表达与临床病理特征的相关性及预后的影响。方法收集胃癌手术切除标本268例,制作组织芯片,采用免疫组化法检测FOXP3在癌周和癌巢的表达,分析FOXP3在癌周及癌巢表达与临床病理特征的相关性及患者生存期的关系,采用双色免疫荧光法检测FOXP3与CD4的共表达情况。结果癌周FOXP3高表达者病死率显著低于低表达者(P=0.01);癌周FOXP3表达与患者的生存期呈正相关(P=0.02)。癌巢FOXP3表达与患者的生存期无相关性;癌周FOXP3表达与CD4表达呈显著的正相关(P=0.001);双色免疫荧光检测显示癌周FOXP3^+细胞与CD4^+细胞存在共表达。结论FOXP3^+/CD4^+免疫细胞多出现在癌周,癌周FOXP3^+细胞可作为预测胃癌患者预后的生物学标志物。
Purpose To analyze the correlation between expression of FOXP3 and clincopathological data or the prognosis of gastric cancer patients.Methods Altogether 268 excised samples were collected from the patients pathologically diagnosed as gastric cancer and made into tissue microarray.The expression patterns of FOXP3+cells in peritumoral and intratumoral tissues were detected on the tissues microarray by immunohistochemical staining.The relationship between expression of FOXP3 and clinicopathologic characteristics,and overall survival time was analysed in all patients.The colocalized expression of FOXP3 and CD4 was detected by a double staining immunofluorescence method.Results The mortality of patients with high peritumoral FOXP3 expression was significantly lower than that of patients with low peritumoral FOXP3 expression(P=0.01).Peritumoral FOXP3 expression was positively correlated with patient survival(P=0.02).The intratumoral FOXP3 expression was no significant correlation with the survival of patients.There was a significant positive correlation between peritumoral FOXP3 expression and CD4 expression(P=0.001).Double staining immunofluorescence detection showed colocalized expression between peritumoral FOXP3 and CD4 positive cells.Conclusion FOXP3 and CD4 positive immunocytes are more likely to appear in peritumor than in intratumor.The peritumoral infiltration FOXP3 cells can be viewed as markers for predicting the prognosis of gastric cancer patients.
