lncRNA LOC285194在肾癌组织中的表达及其对786-O细胞增殖、迁移与侵袭的影响 被引量:1

Expression of lncRNA LOC285194 in renal cell carcinoma and its effect on the proliferation, migration and invasion of 786-O cells

摘要 目的探讨lncRNA LOC285194在肾癌组织中的表达及其对肾透明细胞癌786-O细胞的增殖、迁移与侵袭的影响。方法实时荧光定量聚合酶链反应(qRT-PCR)检测lncRNA LOC285194在28例肾透明细胞癌患者癌组织及相应的癌旁组织、ccRCC细胞株786-O及正常肾上皮细胞系KiMA中的表达。将载有lncRNALOC285194序列的质粒和阴性对照质粒转染ccRCC细胞株786-O,CCK-8实验检测转染后786-O细胞的增殖能力变化,划痕实验检测转染后786-O细胞的迁移能力变化,Transwell法检测转染后786-O细胞的侵袭能力变化,Western blot实验检测转染后786-O细胞周期蛋白D1(cyclinD1)与血管内皮生长因子(VEGF)的表达变化。结果lncRNA LOC285194在肾透明细胞癌组织中的表达明显低于癌旁组织(P<0.01),在ccRCC细胞株786-O中的表达明显低于正常肾上皮细胞系KiMA中的表达(P<0.01)。上调lncRNA LOC285194的表达后,786-O细胞的增殖、迁移与侵袭能力明显降低,cyclin D1与VEGF蛋白的表达明显降低(P<0.01)。结论 lncRNA LOC285194在肾癌组织中低表达,lncRNA LOC285194过表达能够抑制786-O细胞的增殖、迁移与侵袭。 Objective To investigate the expression of lncRNA LOC285194 in renal cell carcinoma and its effect on the proliferation, migration and invasion of 786-O cell line. Methods The expression of lncRNA LOC285194 in 16 cases of clear cell renal cell carcinoma(ccRCC),its adjacent tissues,786-O cells and KiMAcells was detected by realtime fluorescence quantitative polymerase chain reaction(qRT-PCR). The plasmids containing lncrnaloc 285194 sequence and negative control plasmids were transfected into ccrcc cell line 786-O. The proliferation ability of 786-O cells after transfection was detected by CCK-8 experiment. The migration ability of 786-O cells after transfection was detected by scratch experiment. The invasion ability of 786-O cells after transfection was detected by Transwell method. The cyclin D1 and vascular endothelial growth factor(VEGF)of 786-O cells after transfection was detected by Western blot method. Results The expression of lncRNA LOC285194 in renal clear cell carcinoma was significantly lower than that in adjacent tissues(P<0.01), and that in cell line 786-O was significantly lower than that in normal renal epithelial cell line KiMA(P<0.01). After up regulating of the expression of lncRNA LOC285194, the proliferation, migration and invasion ability of 786-O cells was decreased significantly, and the expression of cyclin D1 and VEGF protein was decreased significantly(P<0.01). Conclusion LncrnaLOC285194 is low expression in clear cell renal cell carcinoma, and the overexpression of lncRNA LOC285194 can inhibit the proliferation, migration and invasion of 786-O cells.
