摘要
目的观察Yap对肝癌细胞增殖的影响及可能机制。方法应用Western blot与q RT-PCR方法检测Yap与MALAT1蛋白在肝癌组织及癌旁正常组织中的表达,应用CCK-8法检测si Yap后肝癌细胞增殖活力的改变以及MALAT1的表达变化,过表达MALAT1检测si Yap后肝癌细胞增殖活力的改变。结果 Yap与MALAT1蛋白在肝癌组织中表达水平均高于癌旁正常组织,沉默Yap基因的表达后肝癌细胞增殖活力受到抑制,MALAT1的表达水平减少,过表达MALAT1后阻断Yap表达,Yap对肝癌细胞增殖抑制作用消失。结论 Yap参与肝癌细胞增殖可能与调控MALAT1相关。
Objective To observe the effect of Yap on the proliferation of hepatocellular carcinoma cells and its possible mechanism. Methods Western blot and qRT-PCR were used to detect the expressions of Yap and MALAT1 proteins and mRNAs in hepatocellular carcinoma and adjacent normal tissues, respectively. CCK-8 method was used to detect the viability and proliferation of hepatoma cells. Results The expression levels of both Yap and MALAT1 in hepatocellular carcinoma tissues were significantly increased than those in the adjacent normal tissues. Blocking the expression of Yap could inhibit the cellular proliferation and the expression of MALAT1 in human hepatoma ceils. The ectopic expression of MALAT1 could possibly reverse the inhibitory effects on the cellular viability after Yap knockdown. Conclusions The promotion of Yap on the proliferation of hepatocarcinoma cells might be related to MALAT1.
