摘要
目的基于毒素多肽建立一种新型的全蝎质量检测工艺。方法从东亚钳蝎毒液中分离得到了一种新型钠通道毒素BmK NaTx-13,制备并鉴定了BmK NaTx-13的特异性抗体,并通过竞争性ELISA法检测全蝎提取物中的BmK NaTx-13的含量。结果建立的竞争性酶联免疫吸附法可快速检测全蝎提取物中的BmK NaTx-13的含量并呈现良好的浓度线性依赖。结论该研究为全蝎质控提供了一种快速免疫检测方法,为提升全蝎入药质量标准提供了借鉴。
OBJECTIVE To develop a BmK Scorpion quality control technology based on toxin peptide.METHODS In this study,a novel sodium channel toxin BmK NaTx-13 was isolated from BV,and the specific polyclonal antibody against BmK NaTx-13 was obtained by immunizing rabbits.RESULTS A competitive enzyme linked immunosorbent assay(ELISA)based on BmK NaTx-13 specific antibody could detect the content of BmK NaTx-13 in the processed BmK scorpion extract and showed a good linear concentration dependent.CONCLUSION This research provided a rapid immunoassay method for establishing the quality detection processed BmK scorpion,and provided an insight for improving the quality standard of whole scorpion medicine.
