摘要
目的:探讨大气细颗粒物PM2.5对过敏性鼻炎大鼠临床症状、肺功能、氧化应激水平和免疫功能的影响。方法:75只雄性健康SD大鼠随机分为对照组、模型组、轻度污染组、中度污染组和重度污染组,每组各15只,其中模型组、轻度污染组、中度污染组和重度污染组大鼠建立过敏性鼻炎模型。给予轻度污染组、中度污染组和重度污染组大鼠气管注入1.5 mg/kg、6.0 mg/kg和24.0 mg/kg PM2.5悬液1 mL,给予对照组和模型组大鼠气管注入1 mL生理盐水。所有大鼠均连续染毒处理1个月。分别在建模成功后、染毒完成24 h后记录各组大鼠打喷嚏、搔鼻次数和症状评分。比较各组大鼠染毒完成24 h后血清免疫球蛋白E(IgE)、白细胞介素-4(IL-4)、IL-6、IL-8、IL-10、γ-干扰素(INF-γ)、肿瘤坏死因子(TNF)和丙二醛(MDA)、吸气峰流速(PIFR)、呼气峰流速(PEF)、气道内压(IP)和气道内压最大上升斜率(IP-slope)水平、过氧化物歧化酶(SOD)活性以及肺组织病理学损伤变化情况。结果:轻度污染组、中度污染组和重度污染组大鼠染毒处理后打喷嚏频率、搔鼻频率和症状积分较建模成功时均明显提高(P<0.05),其中重度污染组大鼠染毒处理后打喷嚏频率、搔鼻频率和症状积分明显高于其他两组(P<0.05),中度污染组次之。轻度污染组、中度污染组和重度污染组大鼠染毒处理后MDA水平和SOD活性两两比较,差异有统计学意义(P<0.05),重度污染组大鼠MDA水平最高,SOD活性最低(P<0.05)。中度污染组和重度污染组大鼠IL-4、IL-6、IL-8、IL-10和TNF水平均明显高于其余3组,IFN-γ水平明显低于其余3组(P<0.05),重度污染组大鼠IL-4、IL-6、IL-8、IL-10、TNF和IgE水平水平明显高于中度污染组,IFN-γ水平明显低于中度污染组(P<0.05)。中度污染组和重度污染组大鼠PIFR、PEF均明显低于其他3组,IP和IP-slope均明显高于其他3组(P<0.05)。病理学损伤程度:重度污�
Objective:To investigate the effect of PM2.5 on the clinical symptoms,pulmonary function,oxidative stress level and immune function of allergic rhinitis rats.Methods:75 male healthy SD rats were randomly divided into control group,model group,light pollution group,medium pollution group and heavy pollution group,each with 15 rats,among which model group,light pollution group,medium pollution group and heavy pollution group were used to establish allergic rhinitis model.1 ml of PM2.5 suspension(1.5 mg/kg,6.0 mg/kg and 24.0 mg/kg)was injected into the trachea of rats in the mild pollution group,the moderate pollution group and the severe pollution group,and 1 ml of normal saline was injected into the trachea of rats in the control group and the model group.All rats were exposed for 1 month.The times of sneezing,scratching nose and symptom scores of rats in each group were recorded after the successful modeling and 24 hours after the exposure.The serum levels of Immunoglobulin E(IgE)IL-4,IL-6,IL-8,IL-10,INF-γ,TNF,MDA,peak inspiratory flow rate(PIFR),peak expiratory flow(PEF),intrinsic pressure(IP),IP-slope,the activity of SOD and changes in lung histopathological damage were compared.Results:The sneezing frequency,scratching nose frequency and symptom score of rats in mild pollution group,moderate pollution group and severe pollution group were significantly higher than those in the model group(P<0.05).The sneezing frequency,scratching nose frequency and symptom score of rats in severe pollution group were significantly higher than those in the other two groups(P<0.05),followed by those in moderate pollution group.There was a significant difference in MDA level and SOD activity between the mild pollution group,the moderate pollution group and the severe pollution group(P<0.05).The severe pollution group had the highest MDA level and the lowest SOD activity(P<0.05).The levels of IL-4,IL-6,IL-8,IL-10,and TNF in the moderate and severe pollution groups were significantly higher than those in the other three groups,and
