甘菊提取物对乙醇毒性损伤红细胞变化的影响 被引量:2

摘要 目的探讨甘菊提取物对乙醇致红细胞毒性损伤的保护作用。方法收集2017年9月至2018年5月丽水市人民医院收治急性酒精中毒患者256例和健康体检者85例。均抽取静脉血8 ml,选取乙醇含量为0 mg/100 ml者作为对照组,乙醇含量为80 mg/100 ml者为乙醇毒性损伤组,乙醇含量为80 mg/100ml者加入甘菊提取物(终浓度25、50、100 mg/L)作为不同剂量组,乙醇含量为80 mg/100 ml者加入200 mg/L抗坏血酸作为阳性对照组。分析乙醇对红细胞膜的渗透脆性影响;比较甘菊提取物加入前后对乙醇性红细胞抗氧化酶及丙二醛(MDA)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)巯基(-SH)含量的影响。结果乙醇影响红细胞膜渗透脆性,增加MDA含量,降低红细胞抗氧化酶活性,甘菊提取物能不同程度保护红细胞抗氧化酶活性及增加-SH含量(均P<0.05),显著改善红细胞的形态,其中100mg/L甘菊提取物对红细胞具有保护作用,且与抗坏血酸作用相当。结论甘菊提取物对乙醇致氧化损伤的红细胞具有保护作用,可能是通过对抗氧自由基,抑制脂质过氧化,保护细胞内抗氧化酶活性和细胞膜蛋白上的-SH来实现的。 Objective To investigate the protective effect of chamomile extract on erythrocyte toxic damage induced by ethanol.Methods Peripheral blood samples were collected from 256 alcoholic patients(blood alcohol concentration>80 mg/L)and 85 healthy subjects(blood alcohol concentration=0 mg/L).The erythrocytes were isolated and divided into 6 groups:normal control group,alcohol toxicity group,chamomile low,intermediate and high-dose group(treated with 25,50,100 mg/L of chamomile decoction extract)and AA group(treated with 200 mg/L ascorbic acid).The effects of alcohol on the osmotic fragility of erythrocyte membrane were analyzed.The effects of chamomile extract on the antioxidant enzymes,malonaldehyde(MDA)and sulfhydryl(-SH)content of alcoholic erythrocyte were investigated.Results Ethanol affected the osmotic fragility of erythrocyte membrane,increased MDA content,and decreased the activity of erythrocyte antioxidant enzymes.Chamomile extract protected erythrocyte antioxidant enzyme activity,increased-SH content(P<0.05),and significantly improved the morphology of red blood cells.The protective effect on red blood cells of 100 mg/L chamomile extract was equivalent to that of ascorbic Acid.Conclusion Chamomile extract has protective effects on oxidative damage of red blood cells,which may be achieved by affecting oxygen free radicals,inhibiting lipid peroxidation,protecting intracellular antioxidant enzyme activity and-SH on cell membrane proteins.
