摘要
为研究高压氧(HBO)对缺氧环境中肺癌A549细胞迁移能力的影响及其作用机制,首先用适宜浓度的二氯化钴(CoCl2)模拟A549细胞体内缺氧微环境,再用HBO处理诱导缺氧后的A549细胞,然后分别采取细胞划痕实验、Western Blot法研究不同处理组A549细胞迁移能力及相关蛋白的表达情况。结果表明:在缺氧时A549细胞迁移能力增强,而逆转缺氧状态后迁移能力减弱;缺氧状态时JNK的磷酸化水平增加,而逆转缺氧状态后JNK的磷酸化水平有所降低。说明高压氧能够降低JNK的磷酸化水平,进而使缺氧环境中的A549细胞迁移能力减弱。
Hyperbaric oxygen(HBO)was used to treat lung cancer cells A549 which were exposed to Cobalt dichloride(CoCl 2)to investigate the effect of HBO on the migration of lung cancer cell in hypoxic environment and the possible molecular mechanism.Cell scratch test was used to detect the migration ability of cells A549.Related protein expression was detected by Western Blot.The results show the migration ability of A549 cells increased during hypoxic state,but decreased after treated with HBO.The phosphorylated JNK level is increased in hypoxic state,while the phosphorylated JNK level is decreased after treated with HBO.These suggest that HBO can be used to decrease the phosphorylated JNK level,thus weakening the migration ability of lung cancer cells A549.
