模拟急进高原性脑水肿Wistar大鼠尾状核的1H-MRS分析 被引量:1

Simulated Rapid Progressive Plateau1H-MRS Analysis of Caudate Nucleus in Wistar Rats with Brain Edema

摘要 目的:探讨急进高原性脑水肿(high altitude cerebral edema,HACE)动物模型在模拟高海拔环境中大脑尾状核代谢物的变化,为HACE的诊断寻找更为可靠的检查手段和依据。方法:Wistar大鼠模拟高原低氧环境(7000m),分别在6h、12h、24h、48h、72h五个时间段,每个时间段需6只大鼠,共计30只,为MR扫描组。另选择处于西宁市(平均海拔高度2261m)的大鼠6只为正常对照组。应用动物专用线圈,选择T2WI及尾状核1H-MRS来作为观察大鼠脑水肿后尾状核代谢信息的变化。结果:模拟高原7000m 6h1H-MRS中NAA明显下降、Lac升高,随后NAA逐渐升高,Lac逐渐下降;病理结果显示模拟7000m 6h大鼠脑组织尾状核区细胞水肿、血管扩张,随后细胞肿胀较前减轻;低氧6h Lac/Cr与正常对照组比较差异有统计学意义(P<0.05),低氧48h、72h Lac/Cr与6h比较差异有统计学意义(P<0.05)。结论:1H-MRS能定量检测缺氧环境下脑组织的代谢信息,缺氧早期Lac升高、NAA/Cr下降,1H-MRS有望成为早期缺氧损伤监测、评估预后提供参考指标。 Objective:This paper is to explore the changes of cerebral caudate nucleus metabolite in the animal model of acute high altitude brain edema(high altitude cerebral edema,HACE),and to find more reliable means and basis for the diagnosis of HACE.Methods:At 6 h,12 h,24 h,48 h,72 h in simulated altitude hypoxia environment(7000 m),6 Wistar rats each time point were chosen,respectively,as MR scanning group(n=30).In addition,6 rats at Xining(2261 m)were selected as the normal control group.In this study,T2 WI and caudate hydrogen proton nucleus mono-voxel spectroscopy were used to observe the changes of caudate nucleus metabolic information after brain edema in rats.Results:NAA decreased and LAC increased in1H-MRS at Simulated 7000 m 6 hours,and then NAA gradually increased and LAC gradually decreased,whose pathological results showed edema and vasodilation in caudate nucleus of rats at Simulated 7000 m 6 hours,which then gradually lessened.After 6 hours of Hypoxia,Lac/CR was significantly different from Control Group,and Lac/Cr at 48 and 72 hours of hypoxia was significantly different from 6 Hours group.Conclusion:1H-MRS can quantitatively detect the metabolic information of brain tissue in hypoxia environment.The Lac rises early but NAA/Cr decreases in hypoxia.1H-MRS may be used as an early monitoring index to evaluate the prognosis of hypoxic-ischemic injury.
