308nm准分子光对小鼠湿疹模型表皮及Th细胞因子的影响

Influence of 308 nm Excimer Light on Epidermis and T-helper Cytokines in Eczema Mice Models

摘要 目的观察308 nm准分子光对小鼠湿疹模型皮损及皮损中Th细胞因子的影响,探讨308 nm准分子光治疗湿疹的作用机制。方法采用DNCB外涂法制备小鼠湿疹模型,分为308 nm准分子光治疗组(A组)、莫米松治疗组(B组)、模型组(C组),每组各10只。A组采用隔天1次治疗,连续治疗10次;同期B组外用莫米松乳膏1次/d,至A组治疗结束,共19 d;C组不予处理。比较各组小鼠皮损区表皮厚度变化及白细胞介素(IL)-2、IL-9、IL-10和IL-17表达的差异。结果①308 nm准分子光治疗后,小鼠皮损区表皮厚度较C组明显变薄(F=2.566,P<0.01);B组小鼠皮损改善程度优于A组和C组,差异有统计学意义(F=2.797,7.718,均P<0.01)。②免疫组化染色显示,A组小鼠皮损中IL-9和IL-17表达水平均低于C组(F=1.408,1.459,均P<0.05),IL-2和IL-10表达水平则高于C组(F=1.455,3.542,均P<0.05)。B组小鼠皮损中IL-9和IL-17表达水平均低于C组(F=1.225,1.589,均P<0.01),而IL-2和IL-10表达水平则高于C组(F=1.455,2.667,均P<0.05)。A组和B组之间IL-2、IL-9、IL-10和IL-17的表达差异无统计学意义。结论308 nm准分子光照射小鼠湿疹模型,可改善模型小鼠的皮肤损害;IL-9和IL-17下调、IL-2和IL-10上调,可能与湿疹的发病机制与皮疹转归相关。 Objective In order to observe the influence of 308 nm excimer light on the skin lesions and T-helper cytokines in eczema mice models and explore the mechanism of 308 nm excimer light in the treatment of eczema.Methods Dinitrochlorobenzene(DNCB)was used to establish the eczema mice models,which were randomly divided into 308 nm excimer light treatment group(group A),mometasone treatment group(group B)and model control group(group C),and each group had 10 eczema mice models.Group A was treated once every other day for 10 times,group B was treated once a day for 19 days consecutively,group C was not treated.The changes of epidermis thickness and the expression of interleukin(IL)-2,IL-9,IL-10 and IL-17 were compared.Results①After 308 nm excimer light treatment,the epidermis thickness of the lesions in mice was significantly thinner than that in group C(F=2.566,P<0.01);the improvement of the lesions in mice in group B was better than that in group A and group C(F=2.797,7.718,P<0.01).②The immunohistochemical results showed that the expression levels of IL-9 and IL-17 in group A were lower than those in group C(F=1.408,1.459,P<0.05),while the expression levels of IL-2 and IL-10 were higher than those in group C(F=1.455,3.542,P<0.05).The expression levels of IL-9 and IL-17 in the skin lesions of mice in group B were lower than those in group C(F=1.225,1.589,P<0.01),while the expression levels of IL-2 and IL-10 were higher than those in group C(F=1.455,2.667,P<0.05).There were no significant differences in the expression of IL-2,IL-9,IL-10 and IL-17 between group A and group B.Conclusion 308 nm excimer light irradiation can improve the skin damage of eczema mouse models,the down-regulation of IL-9 and IL-17 and the upregulation of IL-2 and IL-10 may be related to the pathogenesis and prognosis of eczema.
