摘要 目的建立近红外光谱技术快速测定孕马尿中雌酮硫酸钠含量的方法。方法采用SupNIR-2600型近红外光谱仪采集孕马尿的光谱数据,用高效液相色谱法(HPLC)测定110个孕马尿样品中雌酮硫酸钠的含量作为参考值。根据HPLC法含量测定结果,划分为校正集样品88个,验证集22个,结合偏最小二乘法,建立孕马尿中雌酮硫酸钠近红外光谱定量校正模型。结果采用二阶卷积求导的光谱预处理方法,选择1000~1799 cm^-1波段进行孕马尿中雌酮硫酸钠近红外光谱定量校正模型的建立,主成分因子数为3,内部交叉验证决定系数为(R2)为0.9443,校正标准偏差(SEC)为1.77,交互验证校正标准偏差(SECV)为4.16。预测集样品平均相对偏差为1.45%。结论该方法快速、准确、简便,可以用于孕马尿中雌酮硫酸钠含量快速测定。 Objective To establish a method for the rapid determination of sodium estrone sulfate in pregnant mare'′s urine by near infrared spectroscopy.Methods The spectral data of pregnant mare′s urine wascollect byusing SupNIR-2600 near infrared spectrometer.The content of sodium estrone sulfate in 110 pregnantmare′s urine,which was determined byhigh performance liquid chromatography(HPLC)was used as a reference value.According to the content value determined by HPLC method,the sample of pregnant mare′s urinewas divided into 88 calibration sets and 22 validation sets.A near infrared spectroscopy quantitative correction model of sodium estrone sulfate in pregnant mare′s urine was establishedby using partial least squares method.Results By using the second-order convolution-derived spectral pretreatment method,the 1000-1799 cm^-1 band was selected to establish the near infrared spectroscopy quantitative correction model of sodium estrone sulfate in pregnant mare′s urine.Theprincipal component factors was 3 andthe determination coefficient of internal cross-validation(R2)was 0.9443,internal cross-validation the coefficient of determination was 0.9443,the corrected standard deviation(SEC)was 1.77,and the cross-verification corrected standard deviation(SECV)was 4.16.The average relative deviation of the samples in the prediction set is 1.45%.Conclusion This method is fast,accurate and simple.It can be used for the rapid determination of the content of sodium estrone sulfate in pregnant mare′s urine.
