摘要
目的探讨乳腺癌组织长链非编码RNA(lncRNA)牛磺酸上调基因1(TUG1)、组蛋白甲基化转移酶2(EZH2)表达变化及其临床意义。方法选择原发性乳腺癌患者83例,取手术切除的乳腺癌组织及其配对的癌旁正常组织,采用实时荧光定量PCR法检测lncRNA TUG1、EZH2相对表达量。比较乳腺癌组织与癌旁正常组织lncRNA TUG1、EZH2表达,分析乳腺癌组织lncRNA TUG1表达与EZH2表达的关系,以及乳腺癌组织lncRNA TUG1、EZH2表达与患者临床病理特征和预后的关系。结果乳腺癌组织lncRNA TUG1、EZH2相对表达量均高于癌旁正常组织(P均<0.01)。Pearson相关分析显示,乳腺癌组织lncRNA TUG1相对表达量与EZH2相对表达量呈正相关关系(P<0.01)。乳腺癌组织lncRNA TUG1相对表达量与肿瘤TNM分期、淋巴结转移有关(P均<0.05),与患者年龄、乳腺癌家族史、肿瘤直径、组织分化程度无关(P均>0.05);乳腺癌组织EZH2相对表达量与肿瘤组织分化程度、淋巴结转移有关(P均<0.05),与患者年龄、乳腺癌家族史、肿瘤直径、TNM分期无关(P均>0.05)。以乳腺癌组织lncRNA TUG1、EZH2相对表达量的中位数为临界值,将患者分为lncRNA TUG1高表达者46例与lncRNA TUG1低表达者37例、EZH2高表达者43例与EZH2低表达者40例。lncRNA TUG1、EZH2高表达者3年生存率均低于其相应低表达者(P均<0.05)。结论乳腺癌组织lncRNA TUG1、EZH2表达升高,且二者表达呈正相关关系;lncRNA TUG1、EZH2表达升高与乳腺癌恶性进展和患者预后不良有关。
Objective To investigate the changes and clinical significance of long non-coding RNA(lncRNA)taurine up-regulated gene 1(TUG1)and histone methylated transferase 2(EZH2)expression in the breast cancer tissues.Methods Eighty-three patients with primary breast cancer who had received modified radical mastectomy were selected.The surgically resected breast cancer tissues and their paired adjacent normal tissues were collected,and the relative expression levels of lncRNA TUG1 and EZH2 were detected by real-time fluorescence quantitative PCR.The expression levels of lncRNA TUG1 and EZH2 in breast cancer tissues and adjacent normal tissues were compared,the relationship between the expression of lncRNA TUG1 and EZH2 in breast cancer tissues,and the relationship of the expression of lncRNA TUG1 and EZH2 in the breast cancer tissues with the clinicopathological characteristics and prognosis of the patients were analyzed.Results The relative expression levels of lncRNA TUG1 and EZH2 in the breast cancer tissues were higher than those in the adjacent normal tissues(both P<0.01).Pearson correlation analysis showed that the relative expression of lncRNA TUG1 in the breast cancer tissues was positively correlated with the relative expression of EZH2(P<0.01).The relative expression of lncRNA TUG1 in the breast cancer tissues was related to TNM stage and lymph node metastasis(both P<0.05),but not to age,family history of breast cancer,tumor diameter or tissue differentiation degree(all P>0.05).The relative expression level of EZH2 in the breast cancer tissues was related to tissue differentiation degree and lymph node metastasis(all P<0.05),but not to age,family history of breast cancer,tumor diameter or TNM stage(all P>0.05).With the median of relative expression of lncRNA TUG1 and EZH2 in breast cancer tissues as the critical value,there were 46 patients with high expression of lncRNA TUG1,37 patients with low expression of lncRNA TUG1,43 patients with high expression of EZH2,and 40 patients with low expression of EZH2.The 3-y
