摘要
目的研究青蒿素衍生物对卵巢癌细胞体外增殖、凋亡能力的影响及机制。方法实验设置为实验组-1、实验组-2和空白组、对照组,实验组-1、实验组-2卵巢癌OVCAR3细胞分别给予10μmol·L^-1双氢青蒿素及10μmol·L^-1青蒿琥酯处理,对照组和空白组分别给予10μg·mL^-1的顺铂及等量生理盐水处理。以噻唑蓝法检测细胞增殖情况;以Annexin V-FITC/PI双染法流式细胞术检测细胞凋亡情况。结果干预后72 h,实验组-1、实验组-2和对照组细胞增殖抑制率分别为(67.98±3.58)%,(65.15±3.48)%,(61.05±2.49)%,实验组-1、实验组-2增殖抑制率高于对照组(P<0.05)。干预48 h后,实验组-1、实验组-2和对照组、空白组OVCAR细胞早期凋亡率分别为(26.59±4.35)%,(23.14±3.47)%,(19.87±1.36)%,(0.35±0.17)%,晚期凋亡率分别为(19.56±2.36)%,(18.48±2.57)%,(5.72±1.32)%,(0.24±0.05)%。以上指标实验组-1、实验组-2分别与对照组相比差异均有统计学意义(均P<0.05)。结论双氢青蒿素及青蒿琥酯可以抑制卵巢癌OVCAR3细胞增殖,促进其发生凋亡,其机制可能与抑制PTEN-Akt信号通路活性有关。
Objective To investigate the effect and mechanism of artemisinin derivatives on the proliferation and apoptosis of ovarian cancer cells in vitro.Methods The experiment was set up as test group-1,test group-2,blank group and control group.Test group-1,test group-2 were treated with 10μmol·L^-1 dihydroartemisinin and 10μmol·L^-1 artesunate respectively.Control group and blank group were treated with 10μg·mL^-1 cisplatin and the same amount of normal saline respectively.Cell proliferation was detected by methyl thiazolyl tetrazolium(MTT)method,apoptosis rate was detected by Annexin V-FITC/PI double staining flow cytometry.Results After 72 h of intervention the inhibition rates of ovcar cell proliferation in test group-1,test group-2 and control group were(67.98±3.58)%,(65.15±3.48)%,(61.05±2.49)%,and the inhibition rates of test group-1,test group-2 were higher than those in control group(P<0.05).After 48 h of intervention,the early apoptosis rates of cells in test group-1,test group-2,control group and blank group were(26.59±4.35)%,(23.14±3.47)%,(19.87±1.36)%,(0.35±0.17)%,and the late apoptosis rates were(19.56±2.36)%,(18.48±2.57)%,(5.72±1.32)%,(0.24±0.05)%.The difference between test group-1,test group-2 and control group was statistically significant of the above indexes(all P<0.05).Conclusion Dihydroartemisinin and artesunate can inhibit the proliferation and promote the apoptosis of OVCAR3 cells,which may be related to the inhibition of PTEN-Akt signaling pathway.
