茄根醇提物治疗Ⅱ度冻伤模型大鼠实验研究 被引量:4

Experimental Study on the Treatment of Second Degree Frostbite Model Rats with Ethanol Extract of Solanum Melongena Root

摘要 目的观察茄根醇提物对Ⅱ度冻伤SD大鼠表观皮损修复及病理形态、血液流变学的干预作用。方法将15只SPF级SD大鼠按随机数字表法分为生理盐水组、茄根醇提物组、尿素维E乳膏组,每组各5只,液氮法造模,每天2次1mL/100g规格擦涂给药,观察表观皮肤损伤修复、镜下病理形态、血液流变学指标。结果茄根醇提物组、尿素维E乳膏组造模第6天表观指标较生理盐水组显著好转[(-2.20±0.84)、(-2.80±1.30)比(-0.60±0.55),P<0.05],茄根醇提物组与尿素维E乳膏组比较差异无统计学意义(P>0.05)。病理形态示造模第18天茄根醇提物组、尿素维E乳膏组镜下炎细胞较少、已有肉芽组织形成。造模第18天尿素维E乳膏组血液流变学指标血浆黏度[(1.42±0.08)mPa.S比(1.34±0.06)mPa.S,P<0.05]、纤维蛋白原[(3.14±0.17)g/L比(2.94±0.12)g/L,P<0.05]高于生理盐水组。茄根醇提物组较生理盐水组稍有降低但差异无统计学意义(P>0.05)。茄根醇提物组全血黏度切变率[1.00/s、5.00/s、30.00/s、200.00/s:(30.64±7.79)mPa.S比(53.27±16.83)mPa.S、(13.10±2.80)mPa.S比(19.76±4.85)mPa.S、(7.30±1.29)mPa.S比(9.51±1.55)mPa.S、(5.36±0.81)mPa.S比(6.31±0.64)mPa.S,P<0.05]、血浆黏度[(1.34±0.02)mPa.S比(1.42±0.08)mPa.S,P<0.05]、红细胞聚集指数[(5.64±0.68)比(8.30±1.94),P<0.05]、纤维蛋白原[(2.95±0.05)g/L比(3.14±0.17)g/L,P<0.05]均显著低于尿素维E乳膏组。结论茄根醇提物能促进Ⅱ度冻伤SD模型大鼠皮肤修复,促进愈合,作用机制可能与抗炎作用有关。 Objective To observe the intervention effect of ethanol extract of Solanum melongena root on apparent skin lesion repair,pathological morphology and hemorheology in SD rats with second degree frostbite.Methods Fifteen SPF SD rats were randomly divided into normal saline group,ethanol extract of solanaceous root group and urea vitamin E cream group,with 5 animals in each group.The disease model was constructed by liquid nitrogen method,and the rats were rubbed 1 mL/100 g drug twice a day.The apparent skin injury repair,microscopic pathological morphology and hemorheological indexes were observed.Results On the 6 th day of modeling,the apparent indexes of ethanol extract of solanaceous root group and urea vitamin E cream group were significantly better than those of normal saline group[(-2.20±0.837)and(-2.80±1.304)vs(-0.60±0.548),P<0.05,respectively],but there was no significant difference between ethanol extract of solanaceous root group and urea vitamin E cream group.On the 18 th day of modeling,pathological morphology showed that there were fewer inflammatory cells and granulation tissue formation in ethanol extract of solanaceous root group and urea vitamin E cream group;The plasma viscosity of the hemorheologic index[(1.42±0.08)vs(1.34±0.06)mPa.S,P<0.05],and fibrinogen[(3.14±0.17)vs(2.94±0.12)g/L,P<0.05],of urea vitamin E cream group were higher than those of normal saline group.Compared to normal saline group,solanaceous root group decreased slightly,but there was no significant difference.The whole blood viscosity shear rate of 1.00/s[(30.64±7.79)vs(53.27±16.83)mPa.S],5.00/s[(13.10±2.80)vs(19.76±4.85)mPa.S],30.00/s[(7.30±1.29)vs(9.51±1.55)mPa.S]and 200.00/s[(5.36±0.81)vs(6.31±0.64)mPa.S],plasma viscosity[(1.34±0.02)vs(1.42±0.08)mPa.S],red blood cell aggregation index[(5.64±0.68)vs(8.30±1.94)],and fibrinogen[(2.95±0.05)vs(3.14±0.17)g/L]in ethanol extract of solanaceous root group were significantly lower than those in urea vitamin E cream group,P<0.05,respectively.Conclusion Ethanol extra
