定喘汤对哮喘模型小鼠肺组织炎性因子及NLRP3炎症小体的影响 被引量:6

The Effect of Dingchuan Decoction on Inflammatory Factors and Fnflammasome NLRP3 in Lung Tissue of Asthmatic Mice

摘要 目的观察定喘汤对哮喘模型小鼠肺组织炎性因子及NLRP3炎症小体的影响。方法将小鼠随机分为NS+NS组、NS+DCT组、OVA+NS组和OVA+DCT组,每组12只。OVA+NS及OVA+DCT组采取OVA联合LPS致敏复制中性粒细胞为主的哮喘小鼠模型。NS+DCT及NS+NS组采用生理盐水替代。NS+DCT及OVA+DCT组采用定喘汤灌胃;OVA+NS及NS+NS组以生理盐水替代。末次灌胃24 h后,每组取6只小鼠采集BALF,采用ELISA检测IL-1β、IL-4、IL-17的表达;每组另取6只小鼠取左肺中叶行免疫组化检测,取左肺下叶检测IL-1β、NLRP3、caspase-1的蛋白表达水平。结果与NS+NS组和NS+DCT组相比,OVA+NS组和OVA+DCT组小鼠肺组织炎性指标显著上升,肺泡周围出现炎性因子聚集,BLAF中IL-1β、IL-4、IL-17含量显著上升。与OVA+NS组相比,OVA+DCT组BLAF中IL-1β、IL-4、IL-17含量,肺组织炎性指标及NLRP3、caspase-1等蛋白表达均下降。与NS+NS组比较,NS+DCT组小鼠BLAF中IL-1β、IL-4、IL-17含量,肺组织炎性指标及NLRP3、caspase-1等蛋白表达均无显著差异。结论定喘汤可以抑制NLRP3炎症小体,降低IL-1β、IL-17含量,减少炎性因子募集,减轻气道及肺组织中炎症反应和提高抗炎作用,改善气道结构反应,缓解哮喘症状。 Objective To observe the effects of Dingchuan decoction on inflammatory factors and inflammasome NLRP3 in lung tissue of asthmatic mice.Methods Mice were randomly divided into NS+NS group,NS+DCT group,OVA+NS group and OVA+DCT group,with 12 in each group.The OVA+NS and OVA+DCT groups were treated with OVA-LPS to sensitize neutrophil-associated asthma mouse models.The NS+DCT and NS+NS groups were replaced with normal saline(NS).The NS+DCT and OVA+DCT groups were gavaged with Dingchuan decoction.The OVA+NS and NS+NS groups were replaced with NS.After 24 hours of last gavage,six of each group were collected Bronchoalveolar lavage fluid(BALF).The expression of IL-1β,IL-4 and IL-17 was detected by ELISA.Other six of each group were taken from the left middle lobe for immunohistochemistry.Western blot was performed in the left lower lobe to detect the protein expression levels of IL-1β,NLRP3 and caspase-1.Results Compared with NS+NS group and NS+DCT group,the inflammatory index of lung tissue in OVA+NS group and OVA+DCT group increased significantly,inflammatory factor aggregation around alveolar cells,IL-1β,IL-4,IL-17 in BLAF increased significantly.Compared with OVA+NS group,IL-1β,IL-4,IL-17,lung tissue inflammatory index and NLRP3 and caspase-1 were decreased in OVA+DCT group.Compared with NS+NS group,there were no significant differences in IL-1β,IL-4,IL-17,lung tissue inflammatory index and NLRP3,caspase-1 protein expression in NS+DCT group.Conclusion Dingchuan decoction can inhibit inflammasome NLRP3,reduce IL-1β,IL-17,reduce the recruitment of inflammatory factors,reduce inflammation and improve anti-inflammatory effects in airways and lung tissues,improve airway structural response,and alleviate asthma symptoms.
