摘要 目的建立同时测定附子理中丸(大蜜丸)中果糖和葡萄糖含量的方法。方法采用高效液相色谱法(HPLC)示差折光检测器检测,以Prevail Carbohyrate ES为色谱柱,乙腈-水(85:15)为流动相,等度洗脱,流速0.8 ml/min,柱温25℃,进样体积15μl。结果果糖和葡萄糖进样量分别在75.0015~375.0150μg(r=0.9998)、80.0015~400.0075μg(r=1.0000)与峰面积均呈良好线性关系;精密度、稳定性、重复性试验的RSD<2.0%;平均加样回收率分别为100.03%,99.34%(RSD<2.0%,n=9)。结论该方法操作简便、结果准确,可用于附子理中丸(大蜜丸)中葡萄糖和果糖含量的同时测定。 Objective To establish a method for simultaneous determination of glucose and fructose in in Fuzi Lizhong Pills(Big Honeyed Pills) by HPLC. Methods HPLC equipped with differential refractive detector was adopted. The determination was performed on Prevail Carbohyrate ES column with mobile phase consisted of acetonitrile-water(85:15) at the flow rate of 0.8 ml/min, and the column temperature was controlled at 25 ℃. The sample size was 15 μl. Results The concentrations of glucose and fructose showed good linear relationships with the corresponding peak areas in the range of 75.0015-375.0150 μg(r=0.9998) and 80.0015-400.0075 μg(r=1.0000), respecively, and the RSDs of precision, stability and reproducibility tests were all lower than 2.0 %. The average recoveries of fructose and glucose were 100.03 % and 99.34 %(RSD<2.0 %, n=9), respectively. Conclusion The method is simple and accurate, and can be used for simultaneous determination of glucose and fructose in Fuzi Lizhong Pills(Big Honeyed Pills).
