摘要 本试验旨在研究狐源罗伊氏乳杆菌ZJF036发酵特性、黏附能力及动物安全性,探究罗伊氏乳杆菌ZJF036的益生特性。试验检测了罗伊氏乳杆菌ZJF036的生长特性、产酸性能、药物敏感性,对人工胃液、人工肠液和胆盐的耐受性,细菌的疏水率和自凝聚率,对狐狸小肠上皮细胞的黏附能力以及对小鼠血液生理生化指标的影响。结果表明:罗伊氏乳杆菌ZJF036培养4~10 h进入对数生长期;培养菌液的最低pH为3.80;罗伊氏乳杆菌ZJF036对麦迪霉素、头孢曲松和素氯霉敏感,对万古霉素、环丙沙星、丁胺卡那和氨苄西林不敏感;细菌培养上清液对大肠杆菌、沙门氏菌和金黄色葡萄球菌有良好的抑菌作用;罗伊氏乳杆菌ZJF036在人工胃液和人工肠液中3 h存活率可达90%以上,能够耐受0.4%的胆盐环境;罗伊氏乳杆菌ZJF036的疏水率为82.27%,在2和4 h自凝聚率分别为32.47%和41.70%;罗伊氏乳杆菌ZJF036对狐狸小肠上皮细胞的黏附数为(77.0±8.0) CFU/细胞,明显高于鸡源鼠李糖乳杆菌GG;此外,小鼠安全性试验表明,血液生理生化指标均未见异常。综上所述,罗伊氏乳杆菌ZJF036生长迅速,具有良好产酸性能和抑菌能力,可以耐受人工胃液、人工肠液和胆盐,对狐狸小肠上皮细胞具有很强的黏附能力,对小鼠安全可靠。罗伊氏乳杆菌ZJF036可作为犬科动物益生菌制剂的备选菌株。 The objective of this study was to investigate the fermentation characteristics,adhesion ability and animal safety of Lactobacillus reuteri ZJF036 isolated from fox,and to explore the prebiotic property of Lactobacillus reuteri ZJF036.The growth characteristics,acid-producing capacity,drug sensitivity,tolerance to artificial gastric juice,artificial intestinal juice and bile salts,hydrophobicity and self-coacervation rates,adhesion ability to fox intestinal epithelial cells,and mouse blood physiological and biochemical indexes of Lactobacillus reuteri ZJF036 were detected.The results showed that the logarithmic growth phase is 4 to 10 h for bacteria culture of Lactobacillus reuteri ZJF036,the minimum pH of culture medium is 3.80;Lactobacillus reuteri ZJF036 was sensitive to midecamycin,ceftriaxone and chloramphenicol,but not sensitive to vancomycin,ciprofloxacin,amikacin and ampicillin;the supernatant of bacterial culture had good bacteriostatic effects on Escherichia coli,Salmonella and Staphylococcus aureus;the survival rate of Lactobacillus reuteri ZJF036 after culturing with simulated artificial gastric juice and artificial intestinal juice for 3 h was over 90%,and it could tolerate 0.4%bile salt;the hydrophobicity of Lactobacillus reuteri ZJF036 was 82.27%,and the self-coacervation rates at 2 and 4 h were 32.47%and 41.70%,respectively;the adhesion number of Lactobacillus reuteri ZJF036 to fox intestinal epithelial cells was(77.0±8.0)CFU/cell,which was significantly higher than that of Lactobacillus rhamnosus GG isolated from chicken;in addition,the safety test on mouse showed that blood physiological and biochemical indexes were normal.It is concluded that Lactobacillus reuteri ZJF036 grows rapidly,has good acid-producing capacity and bacteriostatic ability,can tolerate artificial gastric juice,artificial intestinal juice and bile salt,has strong adhesion to fox small intestinal epithelial cells,and is safe and reliable to mice.Lactobacillus reuteri ZJF036 is expected to be a candidate strain for preparatio
