紫杉醇联合卡铂放疗对宫颈癌患者血管生成及侵袭能力的影响 被引量:3

Effect of Paclitaxel Combined with Carboplatin on Angiogenesis and Invasive Ability in Patients with Cervical Cancer

摘要 目的探讨紫杉醇联合卡铂同期放疗对宫颈癌患者血管生成及侵袭能力的影响。方法方便选取2017年1月—2019年1月该院收治的宫颈癌患者82例,按随机数字表法分为研究组(n=41,采取紫杉醇联合卡铂同期放射治疗)和对照组(n=41,单纯采取放射治疗),共治疗3个疗程,观察两组临床疗效,治疗前后取病理组织,经免疫组织化学SP法后,在高倍显微镜下观察两组患者治疗前后的微血管密度(MVD)及微淋巴管密度(LVD)变化,采取Western blot法检测两组宫颈癌细胞株Hela细胞中Rock-1、p-cofilin表达,并开展采取Transwell迁移和侵袭实验,读取OD值,观察两组患者细胞体外侵袭能力的变化。结果研究组有效缓解率60.98%高于对照组39.02%,组间比较差异有统计学意义(χ^2=9.644,P<0.05);研究组治疗后MVD、LVD值分别为(9.83±1.14)个/高倍视野、(2.24±0.29)个/高倍视野,均低于对照组,组间比较差异有统计学意义(t=12.678、23.549,P<0.05);研究组Rock-1、p-codinlin蛋白表达及OD值均低于对照组,组间比较差异有统计学意义(t=9.545、5.587,P<0.05)。结论对宫颈癌患者采取紫杉醇联合卡铂同期放疗治疗可取得良好效果,抑制肿瘤血管的生成,同时减少细胞侵袭能力。 Objective To investigate the effect of concurrent radiotherapy with paclitaxel and carboplatin on angiogenesis and invasive ability in patients with cervical cancer.Methods Eighty-two patients with cervical cancer treated in our hospital from January 2017 to January 2019 were convenient randomly divided into a study group(n=41,paclitaxel combined with carboplatin concurrent radiation therapy)and a control group(n=41,taking radiotherapy alone),a total of 3 courses of treatment,to observe the clinical efficacy of the two groups,pathological tissues were taken before and after treatment,and after treatment with immunohistochemical SP method,the microvessel density before and after treatment was observed under high power microscope(MVD)and microlymphatic vessel density(LVD)changes.Western blot was used to detect the expression of Rock-1 and p-cofilin in two groups of cervical cancer cell lines Hela cells.Transwell migration and invasion experiments were performed to read the OD value and observe changes of invasion ability of cells in two groups of patients in vitro.Results The effective response rate of the study group was 60.98%higher than that of the control group 39.02%.There was a statistical difference between the groups(χ^2=9.644,P<0.05).The MVD and LVD values of the observation group after treatment were(9.83±1.14)/high visual fields,(2.24±0.29)/high magnification fields were lower than those in the control group.There was a statistical signifant difference between the groups(t=12.678,23.549,P<0.05);Rock-1,p-codinlin protein expression and OD values in the study group were lower than those in the control group,and there were statistical differences between the groups(t=9.545,5.587,P<0.05).Conclusion Concurrent radiotherapy of paclitaxel combined with carboplatin for cervical cancer patients can achieve good results,inhibit tumor angiogenesis,and reduce cell invasion.
