摘要
目的:通过饮食和运动干预,探讨饮食和运动对不同体脂雄性小鼠氧化应激和精子质量参数的影响,为揭示饮食和运动改善生殖功能提供依据。方法:36只C57BL/6J雄性小鼠随机分成标准饲料组和高脂饲料肥胖造模组,标准饲料组即为标准饲料对照组(NC,n=8)。饲养12周时,选取高脂饲料肥胖造模成功的小鼠,然后按照体重分层抽样分成3组:肥胖对照组(HC,N=8)、饮食干预组(HN,n=8)、饮食和运动干预组(HNE,n=9)。运动组进行8周跑台运动,每周休息1天,末次运动后36h取材,测量小鼠体重、腹腔脂肪重量、睾丸重量、睾丸系数、血清睾酮,睾丸MDA和SOD,显微镜下测定精子总数和精子活动度。结果:(1)与NC组相比,HC组小鼠的体重、腹腔脂肪重量、lee’s指数均显著性升高(P<0.01),MDA显著性升高(P<0.01),SOD、SOD/MDA均显著性降低(P<0.01),睾丸系数、血清睾酮均显著性下降(P<0.01),精子总数与精子活动度均显著性降低(P<0.05,P<0.01);(2)与HC组相比,HN组小鼠的体重、腹腔脂肪重量、lee’s指数均显著性下降(P<0.01),MDA显著性下降(P<0.01),SOD活力、SOD/MDA均显著性升高(P<0.01),睾丸系数显著性上升(P<0.05),血清睾酮、精子总数和精子活动度无显著性差异;HNE组小鼠的体重、腹腔脂肪重量、lee’s指数均显著性下降(P<0.01),MDA显著性下降(P<0.01),SOD活力、SOD/MDA显著性升(P<0.01),睾丸系数、血清睾酮均显著性上升(P<0.01),精子总数和精子活动度显著性升高(P<0.05,P<0.01);(3)与HN组相比,HNE组小鼠的腹腔脂肪重量、lee’s指数均显著性下降(P<0.01),体重无显著性差异,MDA显著性下降(P<0.01),SOD无显著性差异,SOD/MDA显著性升高(P<0.05),睾丸系数无显著性差异,血清睾酮显著性升(P<0.01),精子活动度显著性升(P<0.05)。结论:(1)长期高脂饮食会引起小鼠体重和腹腔脂肪重量增加,使机体处于过度氧化应激状态,诱使睾丸系数、血清睾酮和精子质量参�
Objective: Through diet and exercise intervention, to explore the effects of diet and exercise on oxidative stress and sperm quality parameters in male mice with different body fat, and to provide an experimental basis for revealing diet and exercise to improve reproductive function. Methods:36 C57 BL/6 J male mice were randomly divided into two groups: the standard feed group and the high-fat diet obesity model group. The standard feed group was the standard feed control group(NC, N=8). After 12 weeks of feeding, the mice with high-fat diet obesity model were selected and divided into three groups according to their body weight: the obesity control group(HC, N=8), the diet intervention group(HN, N=8), the diet and exercise intervention. Group B(HNE, N = 9). In the exercise group, 8 weeks treadmill exercise was performed, 6 days/w, 60 minutes/time, one day of rest per week,36 hours after the last exercise. The body weight, abdominal fat weight, testicular weight, testicular coefficient,serum testosterone, testicular MDA, and SOD were measured, and the total number of sperm and sperm motility was measured under the microscope. Results:(1)Compared with NC group, the body weight, abdominal fat weight and lee’s index of mice in HC group were significantly increased(P<0.01), MDA were significantly increased(P<0.01), SOD and SOD/MDA were significantly decreased(P<0.01),testosterone coefficient and serum testosterone were significantly decreased(P< 0.01), sperm count and activity were significantly decreased(P<0.05, P<0.01).(2)Compared with HC group, the body weight, abdominal fat weight and lee’s index of mice in HN group were significantly decreased(P<0.01), MDA were significantly decreased(P<0.01), SOD activity and SOD/MDA were significantly increased(P<0.01), testicular coefficient were significantly increased(P<0.05), serum testosterone,sperm count and activity had no significant difference.
