摘要
采用乙醇提取法外加超声波辅助,探究不同条件下黑枸杞提取液对常见致病菌(包括大肠埃希氏菌E.coli、金黄色葡萄球菌S.au、肠炎沙门氏菌S.en)的抑菌效果,并在最佳抑菌效果的提取条件下,采用DPPH法测定提取液的抗氧化能力。结果表明:超声时间30 min,超声频率60 Hz,浸提温度25℃,料液比1∶15(g∶mL),乙醇浓度60%时,提取液抑菌性能最佳。尤其对S.en抑菌效果最好,最低抑菌浓度为0.125 g/mL。在上述提取条件下,黑枸杞提取液对自由基的清除率随浓度的增大而显著增大,具有明显的量效关系,溶液浓度在600μg/mL以上时,黑枸杞提取物对自由基的清除率高于同浓度下的VC水溶液,最大清除率可达76.26%。说明黑枸杞浸提液具有明显的抑菌效果和较强的抗氧化性。
Using ethanol extraction and ultrasonic assisted methods to explore the antibacterial effects of Lycium ruthenicum Murr.extracts under different conditions on common pathogens[including Escherichia coli(E.coli),Staphylococcus aureus(S.au),and Salmonella enteritidis(S.en)].Under the best antibacterial effect,the DPPH method is used to determine the antioxidant capacity of the extract.The results showed that when the ultrasonic time was 30 min,the ultrasonic frequency was 60 Hz,the extraction temperature was 25℃,the ratio of material to liquid was 1∶15(g∶mL),and the ethanol concentration was 60%,the extract had the best antibacterial performance.In particular,it had the best antibacterial effect on S.en,with a minimum inhibitory concentration of 0.125 g/mL.Under the above extraction conditions,the scavenging rate of L.ruthenicum on free radicals increased significantly with the increase of the concentration,which had an obvious effect and dose relationship.When the solution concentration was above 600μg/mL,the scavenging rate of L.ruthenicum extract on free radicals was higher than that of the Vc aqueous solution at the same concentration,and the maximum scavenging rate can reach 76.26%.It shows that the L.ruthenicum extract has obvious antibacterial effect and strong anti-oxidation.
