摘要
目的:分离6年生人参内生细菌,并初步分析其代谢产物粗提物体外抗肿瘤活性。方法:采用75%乙醇、次氯酸钠联合应用的方法消毒6年生人参,分离获得其中内生细菌,16SrRNA测序确定内生细菌的种属,利用MTT方法检测内生细菌发酵代谢产物抗肿瘤活性。结果:从6年生人参中分离得到内生细菌18株,其中有3株内生细菌代谢产物粗提物对人宫颈癌细胞Hela的IC50分别为8.4μg/ml、10.3μg/ml、11.6μg/ml,具有较好的抗肿瘤活性。在倒置光学显微镜下,可以观察到明显的肿瘤细胞皱缩等凋亡现象。结论:人参中含有丰富的内生菌资源,且其中含有抗肿瘤活性物质,这为寻找抗肿瘤药物提供了新的思路和研究方向。
Objective: To isolate endophytic bacteria from six - year - old Panax ginseng Meyer and preliminarily analyze the anti- tumor activity of crude extract of its metabolites. Methods: The six- year- old Panax ginseng Meyer was disinfected by 75% ethanol and sodium hypochlorite, the endophytic bacteria were isolated and obtained, 16SrRNA sequencing was used to determine the species, MTT as- say was used to detect the antitumor activity of metabolites. Results : Eighteen endophytie bacteria were isolated from six - year - old Panax ginseng Meyer. IC50 values of crude extracts of metabolites of three strains of Panax ginseng Meyer to human cervical can HeLa cells were 8.4 μg/ml, 10. 3 μg/ml and 11.6 μg,/ml, respectively, the antitumor activity was good; apoptosis of HeLa cells could be observed under inverted optical microscope, such as shrinkage of HeLa cells. Conclusion: Panax ginseng Meyer is rich in endophytic bacteria, and it con- tains antitumor active substances, which provide new ideas and research directions for finding anticancer drugs.
