摘要
目的研究高压氧(HBO)干预对大鼠脊髓损伤(SCI)区水肿及凋亡的影响。方法选取成年雌性SD大鼠60只,随机分为假手术组、SCI组和HBO组各20只,造模后7d,通过TUNEL法检测神经细胞的凋亡情况,以RT-PCR、Westernblot法检测SCI区MMP9/2基因和蛋白的表达。通过HE染色观察SCI区病理组织形态学变化及水肿情况。结果造模后7d,假手术组凋亡细胞数为(6.03±2.34),HBO组凋亡细胞数为(37.94±4.72),均明显低于SCI组的(58.47±5.98);MMP9/2基因表达水平,假手术组分别为[(0.55±0.09),(1.03±0.15)],HBO组为[(1.02±0.16),(1.65±0.28],均明显低于SCI组的[(1.82±0.35),(2.35±0.46)];MMP9/2蛋白表达水平,假手术组分别为[(0.86±0.16),(1.13±0.24)],HBO组为[(1.22±0.22),(1.76±0.34)],均明显低于SCI组的[(1.98±0.37),(2.52±0.64)],差异均有统计学意义(P<0.05)。假手术组大鼠脊髓SCI区未见脊髓水肿。SCI组大鼠SCI区组织水肿明显,HBO组大鼠SCI区组织水肿程度减轻。结论HBO治疗通过减少大鼠SCI区神经细胞的凋亡和减轻SCI区水肿,对脊髓损伤起到了保护作用。
Objective To study the effect of hyperbaric oxygen(HBO)intervention on edema and apoptosis in spinal cord injury(SCI)area of rats.Methods Sixty adult female SD rats were selected and randomly divided into sham operation group,SCI group,and HBO group,with 20 rats in each group.Seven days after the establishment of the model,apoptosis of nerve cells were detected by the TUNEL method.The expressions of MMP9/2 gene and protein in SCI area were detected by RT-PCR and Western blot.The histological changes and edema of the SCI area were observed by HE staining.Results Seven days after modeling,the number of apoptotic cells in sham operation group and HBO group were(6.03±2.34)and(37.94±4.72),respectively,which were significantly lower than that in SCI group with(58.47±5.98);the levels of MMP9/2 gene expression in the sham operation group were[(0.55±0.09),(1.03±0.15)],and the HBO group were[(1.02±0.16),(1.65±0.28)],which were significantly lower than those in the SCI group with[(1.82±0.35),(2.35±0.46)];the levels of MMP9/2 protein expression in the sham operation group were[(0.86±0.16),(1.13±0.24)],and the HBO group were[(1.22±0.22),(1.76±0.34)],which were significantly lower than those in the SCI group with[(1.98±0.37),(2.52±0.64)],and the differences were statistically significant(P<0.05).There was no spinal cord edema in SCI area of sham operated rats.The tissue edema in SCI area of rats in SCI group was obvious,while in HBO group the degree of tissue edema in SCI area decreased.Conclusion HBO treatment could protect SCI by reducing the apoptosis of neurons in SCI area and the edema in SCI area.
