14-3-3σ蛋白在TCDD致胎鼠腭裂中的作用机制的初步研究

摘要 目的:研究14-3-3σ蛋白在2, 3, 7, 8-四氯二苯二噁英(TCDD)致胎鼠腭裂中的作用机制。方法:胚胎日第10.5天(E10.5)时,TCDD组按28μg/kg给予孕鼠一次性灌胃,对照组为等量玉米油灌胃。分别在E13.5、E14.5及E15.5取出胎鼠腭突组织和胎头。免疫组化检测14-3-3σ蛋白表达分布,RT-QPCR和WB检测胎鼠腭突组织14-3-3σ的mRNA和蛋白表达量,P53蛋白和磷酸化P53蛋白表达量。结果:14-3-3σ蛋白主要表达在腭上皮细胞。与对照组比较,TCDD组14-3-3σmRNA和蛋白表达升高;E13.5 TCDD组P53蛋白表达升高,其磷酸化水平在E13.5和E14.5升高。结论:TCDD引起腭突组织14-3-3σ蛋白表达升高,激活P53通路异常,使腭中缝上皮持续存在,干扰腭发育的正常进程。 Objective: To investigate the mechanism of 14-3-3σ protein in the development of cleft palate by maternal exposure to 2,3,7,8-Tetrachlorodibenzo-p-dioxin(TCDD) in fatal mice. Methods: On E10.5, the mice in TCDD group were orally administrated with TCDD 28 μg/kg, while the mice in control group received equivalent corn oil. The pregnant mice in each group were sacrificed on E13.5, E14.5 and E15.5 respectively, fetal palates and heads were collected for analysis. 14-3-3σ protein was detected by immunohistochemical staining. 14-3-3σ mRNA in palate was evaluated by RT-qPCR. Protein of 14-3-3σ,P53 and phosphorylated P53 were detected by Western blot. Results: 14-3-3σ protein was mainly expressed in palatal epithelial cells. The expression of 14-3-3σ mRNA and protein in TCDD group was higher than that in control group(P<0.05);on E13.5, the expression of P53 protein in TCDD group was higher than that in control group;on E13.5 and E14.5, the expressions of phosphorylated P53 protein increased in TCDD group(P<0.05). Conclusion: Elevated expression of 14-3-3σ protein by TCDD may causes medial edge epithelium persistent existence and interferes with the normal development process of the palate through P53 pathway.
