摘要
目的:预测真武汤治疗慢性心力衰竭(chronic heart failure,CHF)的作用机制。方法:运用中药系统药理学分析平台数据库获取真武汤的主要化学成分及靶标基因,CHF的靶标基因从Gene Card数据库收集,取二者交集构建化合物-靶点网络,网络可视化结果分析通过Cytoscape 3.7.1软件展示,蛋白质间相互作用网络构建使用STRING操作并筛选出核心网络,借助DAVID数据库进行基因本体功能(gene ontology,GO)分析和基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)通路富集分析,从而探究真武汤治疗CHF的作用机制。结果:检索到真武汤的59个化合物,其中15个靶点与CHF相互关联。GO富集分析得到24条相关条目,真武汤可能是通过调节各种蛋白受体活性、内切酶活性、胆碱能受体活性发挥治疗作用;KEGG富集分析结果表明真武汤共涉及了110条信号通路,表明真武汤可能通过凋亡信号通路、P53信号通路、PI3K-Akt信号通路、NF-kappaβ信号通路、TNF信号通路等干预细胞凋亡、抑制促炎因子的基因表达从而达到治疗CHF的作用。结论:本研究预测结果表明,真武汤对CHF的防治作用为多成分、多靶点、多通路、多选择的复杂过程,且主要通过影响细胞凋亡通路等代谢途径起效。
Objective:To predict the mechanism of Zhenwu Tang in treatment of chronic heart failure(CHF).Methods:The main chemistries and target genes of Zhenwu Decoction were obtained using the database of Chinese Medicine System Pharmacology Analysis Platform.The target genes of CHF were collected from the Gene Card database.The intersection of the two was used to construct a compound-target network.The network visualization results were analyzed through Cytoscape 3.7.1 Software display,protein interaction network construction uses STRING operation and screens out the core network,with the help of DAVID database for gene ontology(GO)analysis and genomic encyclopedia(Kyoto Encyclopedia of Genes and Genomes,KEGG)pathway enrichment Analysis to explore the mechanism of Zhenwu Decoction in the treatment of CHF.Results:59 compounds of Zhenwu Decoction were retrieved,15 of which were related to CHF.GO enrichment analysis resulted in 24 related entries.Zhenwu Decoction may play a therapeutic role by regulating the activity of various protein receptors,endonuclease activity,and cholinergic receptor activity;KEGG enrichment analysis results showed that Zhenwu decoction involved a total of 110 Signal pathways,indicating that Zhenwu Decoction may be able to achieve treatment by interfering with apoptosis,inhibiting gene expression of proinflammatory factors through apoptosis signaling pathway,P53 signaling pathway,PI3 K-Akt signaling pathway,NF-kappa beta signaling pathway,TNF signaling pathway,etc.,so as to play the role of treating CHF.Conclusion:The prediction results of this study indicate that the prevention and treatment effect of Zhenwu Decoction on CHF is a complex process of multi-component,multi-target,multi-pathway,and multi-selection,and it mainly works by affecting metabolic pathways such as apoptosis pathway.
