辣木叶不同极性部位抗氧化活性的研究 被引量:4

摘要 目的:测定辣木叶醇提物不同极性部位总黄酮含量及抗氧化活性。方法:以石油醚、乙酸乙酯、正丁醇依次萃取辣木叶醇提物,得到不同极性部位并测定各部位总黄酮含量;以Vc为阳性对照,比较辣木叶醇提物及石油醚萃取物、乙酸乙酯萃取物、正丁醇萃取物、水溶性部位对DPPH·、ABTS+·、OH·自由基的清除率和还原能力大小。结果:辣木叶乙酸乙酯部位总黄酮含量明显高于其他部位,达到(326.19±2.35)mg/g;辣木叶醇提物及不同极性部位均有一定的抗氧化作用,其抗氧化活性顺序为:乙酸乙酯部位>正丁醇部位>醇提物>水溶性部位>石油醚部位,其中,当乙酸乙酯部位浓度为0.10 mg/mL时,对DPPH·、ABTS+·、·OH的清除率达到(94.31±2.31)%、(98.43±2.27)%、(74.69±2.17)%,其相应的IC50值为0.011、0.006、0.378 mg/mL。结论:辣木叶醇提物乙酸乙酯部位中总黄酮含量最高且抗氧化活性最强,为辣木叶资源的深入研究和开发利用提供了科学依据。 Objective:To determine the total flavonoids content and antioxidant activity of different polar extracts of ethanol extract of Moringa oleifera leaves.Methods:The extracts of Moringa oleifera leaves were extracted with petroleum ether,ethyl acetate and n-butanol to obtain different polarextracts and to determine the content of total flavonoids in different polar extracts.Using Vc as positive control,the reducing capacity and scavenging rate of ethanol extract,petroleum ether extract,ethyl acetate extract,n-butanol extract and water-soluble extract of Moringa oleifera leaves against 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH·),2,2’-diazo-bis(3-ethylbenzothiazole-6-sulfonic acid)diammonium salt(ABTS+·),hydroxyl radical(OH·)were compared.Results:The total flavonoid content in the ethyl acetate extract of Moringa oleifera leaves was significantly higher than that in other extracts,reaching(326.19±2.35)mg/g.The ethanol extract of Moringa oleifera leaves and its polar extracts all had certain antioxidant activity.The antioxidant activity order was:ethyl acetate extract>n-butanol extract>total ethanol extract>water-soluble extract>petroleum ether extract.When the concentration of ethyl acetate extract was 0.10 mg/mL,the scavenging rate of DPPH·,ABTS+·and OH·reached(94.31±2.31)%,(98.43±2.27)%,(74.69±2.1)%,the corresponding IC50 values were 0.011 mg/mL,0.006 mg/mL and 0.378 mg/mL,respectively.Conclusion:The ethyl acetate extract of Moringa oleifera leaves has the highest content of total flavonoids and the strongest antioxidant activity,which provides a scientific basis for further research and development and utilization of Moringa oleifera leaves resources.
