摘要
目的 观察青海高原地区藏族宫颈癌新辅助化疗敏感性相关miRNA表达谱,探讨其与新辅助化疗疗效及敏感性之间的关系.方法 收集青海地区藏族宫颈鱗癌患者36例,采用实时定量PCR法检测患者化疗前后血清中miR-96、miR-138及miR-720表达情况.结果 与化疗前相比,治疗敏感组化疗后miR-138和miR-720表达均上调,miR-96表达明显下调(P<0.05);治疗抗拒组化疗前后miR-96、miR-138及miR-720表达无明显差异(P>0.05).经Spearman分析,化疗前后miR-96表达变化与肿瘤体积变化呈正相关,miR-138及miR-720表达变化与肿瘤体积变化呈负相关(P<0.05).结论 miR-96、miR-138及miR-720与宫颈癌化疗敏感性相关,可为宫颈癌新辅助化疗敏感性指标提供依据,易于青海高原地区藏族宫颈癌患者的个体化治疗.
Objective To observe the expression profiles of microRNAs related to neoadjuvant chemotherapy sensitivity in Tibetan cervical cancer in Qinghai plateau,and to explore the relationship between the expression profiles and the efficacy and sensitivity of neoadjuvant chemotherapy.Methods Thirty-six Tibetan patients with cervical squamous cell carcinoma in Qinghai were collected.The expression of microRNA-96,microRNA-138 and microRNA-720 in serum of patients before and after chemotherapy were detected by real-time quantitative PCR.Results Compared with before chemotherapy,the expressions of microRNA-138 and microRNA-720 were up-regulated and the expression of microRNA-96 was down-regulated in the treatment sensitive group(P<0.05),while the expressions of microRNA-96,microRNA-138 and microRNA-720 were not significantly different in the treatment resistant group(P>0.05).Spearman analysis showed that the expression of microRNA-96 was positively correlated with the change of tumor volume before and after chemotherapy,and the expression of microRNA-138 and microRNA-720 was negatively correlated with the change of tumor volume(P<0.05).Conclusion Mi-96,miR-138 and mir-720 are correlated with the chemosensitivity of cervical cancer,which can provide evidence for the neoadjuvant chemosensitivity index of cervical cancer and facilitate the individualized treatment of Tibetan cervical cancer patients in Qinghai Plateau.
