摘要
【目的】观察黄芪桂枝五物汤加味对变应性鼻炎(AR)大鼠的治疗作用及可能机制。【方法】将60只Wistar大鼠随机分为正常组、模型组、中药组和氯雷他定组。除正常组,其他组别大鼠采用卵清蛋白诱导AR模型。造模成功后,中药组给予黄芪桂枝五物汤加味灌胃,氯雷他定组给予氯雷他定生理盐水溶液灌胃,正常组、模型组给予等容积生理盐水灌胃,共28 d。统计鼻炎症状评分,酶联免疫吸附分析(ELISA)检测血清黏蛋白5AC(MUC5AC)、黏蛋白5B(MUC5B)、免疫球蛋白E(IgE)、组胺、白三烯、干扰素γ(IFN-γ)、白细胞介素2(IL-2)、白细胞介素4(IL-4)、白细胞介素5(IL-5)、白细胞介素17(IL-17)、白细胞介素10(IL-10)的水平。【结果】与正常组比较,模型组大鼠血清IFN-γ、IL-2、IL-10水平明显降低(P<0.05),IL-4、IL-5、IL-17、MUC5AC、MUC5B、IgE、组胺、白三烯水平和鼻炎症状评分明显升高(均P<0.05);与模型组比较,中药组和氯雷他定组大鼠血清IFN-γ、IL-2、IL-10水平明显升高(均P<0.05),IL-4、IL-5、IL-17、MUC5AC、MUC5B、IgE、组胺、白三烯水平和鼻炎症状评分明显降低(均P<0.05),且2个治疗组比较,差异无统计学意义(P>0.05)。【结论】黄芪桂枝五物汤加味可以通过调节Th1/Th2和Th17/Treg细胞免疫平衡改善AR大鼠症状。
Objective To observe the therapeutic effect of Modified Huangqi Guizhi Wuwu Decoction for rats with allergic rhinitis and its mechanism.Methods Sixty Wistar rats were randomly divided into normal group,model group,Chinese medicine group and loratadine group.Apart from the normal group,the rat model of allergic rhinitis was established by ovalbumin induction in the other groups.After successful modeling,the Chinese medicine group was given intragastrical administration of modified Huangqi Guizhi Wuwu Decoction,the loratadine group was intragastrical administration of loratadine normal saline solution,and the normal group and model group were given intragastrical administration of normal saline,the treatment lasting 28 days.The scores of rhinitis symptoms were calculated,the levels of mucin 5AC(MUC5AC),mucin 5B(MUC5B),immunoglobulin E(IgE),histamine,leukotrienes,interferon-γ(IFN-γ),and interleukin(IL)-2,IL-4,IL-5,IL-17,IL-10 were detected by enzyme-linked immunosorbent assay(ELISA).Results Compared with the normal group,the levels of serum IFN-γ,IL-2 and IL-10 in the model group were significantly decreased(all P<0.05),and the levels of serum IL-4,IL-5,IL-17,MUC5AC,MUC5B,IgE,histamine,leukotrienes and the scores of rhinitis symptoms were increased significantly(all P<0.05).Compared with the model group,the levels of serum IFN-γ,IL-2 and IL-10 were significantly increased,and the levels of serum IL-4,IL-5,IL-17,MUC5AC,MUC5B,IgE,histamine,leukotrienes and the scores of rhinitis symptoms were significantly decreased in the Chinese medicine group and loratadine group(all P < 0.05),and there was no significant difference in the twotreatment groups(P > 0.05). Conclusion Modified Huangqi Guizhi Wuwu Decoction has effects on improving thesymptoms of AR rats by regulating the immune balance of Th1/Th2 and Th17/Treg cells.
