摘要
为探明冠突散囊菌分生孢子的最佳产孢工艺,选取PDL液体为基础培养基,探讨不同的蔗糖浓度、NaCl浓度、培养温度和发酵时间对冠突散囊菌分生孢子产量的影响,采用单因素试验和正交试验对发酵条件进行优化.研究结果表明:当PDL液体培养基的蔗糖含量为20%时,冠突散囊菌分生孢子的最佳产孢工艺条件为NaCl含量10%、培养温度34℃和培养时间5 d;对冠突散囊菌两性型表观形态及显微特征的差异进行了分析,发现冠突散囊菌的有性型与无性型形态差异较为显著.
In order to explore the best sporulation technology of conidia of Eurotium cristatum, the PDL liquid medium is selected as the base medium, the effects of different sucrose concentration, NaCl concentration, culture temperature and fermentation time on spore production of the conidia of the Eurotium cristatum are investigate. Single factor experiment and orthogonal experiment are used to optimize the fermentation conditions. The results show that when the content of sucrose in the PDL liquid medium is 20%, the best process conditions for the spore production of the Eurotium cristatum are the content of NaCl is 10%, the culture temperature is 34 ℃, and the fermentation time is 5 days. By analyzing the differences between the morphological and microscopic characteristics of the Eurotium coronarium, it is found that the differences between the sexual and asexual types of the Eurotium coronarium are significant.
