摘要
目的探索胡桃醌导致小鼠SH-SY5Y神经母细胞瘤凋亡的机理。方法采用转录组技术,对胡桃醌干预小鼠SH-SY5Y神经母细胞瘤后的差异基因进行分析,并选择其中显著性基因,围绕其信号通路分析促凋亡机理,并采用Westem-blot 及RT-PCR等实验方法进行验证。结果采用 Rwith edgeR package进行差异基因分析,共找到821个差异基因。通过KEGG注释,确定了p53信号通路,实验验证表明,不同浓度胡桃醌诱导SH-SY5Y细胞凋亡后p53基因及mRNA表达水平明显增强,其表达量随着胡桃醌浓度的升高而升高。结论胡桃醌导致SH-SY5Y神经母细胞凋亡的机制可能是胡桃醌能明显上调p53 mRNA及p53的表达,从而诱导小鼠SH-SY5Y神经母细胞凋亡。
Objective To explore the mechanism of Juglone induced apoptosis in mouse SH-SY5Y neuroblastoma.Methods The differentially expressed genes of SH-SY5Y neuroblastoma in mice were analyzed by transcriptome technique,and the significant genes were selected.The proapoptotic mechanism was analyzed around its signaling pathway,and Western-blot was used.Experimental methods such as RT-PCR were performed.Results Differential genetic analysis was performed using the Rwith edgeR package to find 821 differential genes.The p53 signaling pathway was confirmed by KEGG annotation.The experimental results showed that the expression of p53 gene and mRNA was significantly increased after the apoptosis of SH-SY5Y cells induced by different concentrations of Juglone,and its expression increased with the increase of the concentration of Juglone.Conclusion The mechanism of the apoptosis of SH-SY5Y neuroblasts by Juglone can be that the expression of p53 mRNA and p53 can be up-regulated by Juglone.
