摘要
为明确高粱黑粉菌中黑色素的开发价值,采用盐酸水解和氢氧化钠碱溶液提取的方法制备高粱黑粉菌黑色素,研究其理化性质以及稳定性。采用紫外分光光度计和红外光谱仪对高粱黑粉菌黑色素进行检测,并且应用红外光谱仪考察不同pH值的条件下高粱黑粉菌黑色素同Cu^2+和Fe^3+的相互作用的情况。结果表明:高粱黑粉菌黑色素的最大吸收波长在215nm;在pH值2.0~5.0的范围内呈现浅棕色;pH值7.0为时为棕红色;当pH值大于8.0时,高粱黑粉菌黑色素的颜色变为深棕色;除还原剂以外,热、光、微波、氧化剂及常用食品添加剂对其无影响,金属离子Cu^2+、Mg^2+、Ca^2+、Zn^2+、A1^3+和Fe^3+对高粱黑粉菌黑色素的稳定性都没有不良影响。红外光谱表明高粱黑粉菌黑色素含有OH、N-H、C=O、C=C、羧基、吡喃环、醇羟基和酚羟基。另外,还揭示Cu^2+和Fe^3+主要与高粱黑粉菌黑色素中的羧基和酚羟基发生了络合作用。这些结果说明,高粱黑粉菌黑色素具有良好的稳定性,是一种非常具有开发应用前景的天然色素,为其作为功能食品的进一步加工利用开发提供理论依据和方法。
In order to determine the development value of melanins from Sphacelotheca reiliana,the physic-chemical properties were examined.In this research,the melanin was prepared by the HCl hydrolysis and NaOH leaching solution extraction,and its physicochemical properties and stability were studied.Ultraviolet spectrophotometer and infrared spectrometer were used to detect the melanins.Further,the interactions of melanins with Cu^2+ion and Fe^3+ion were investigated by IR spectroscopy under different pH values.The results showed that the maximum absorption wavelength of melanin was 215nm,the color of the melanin has a light brown in the pH range of 2.0-5.0,brownish red when pH 7.0,and turns to dark brown when pH>8.0;in addition to reducing agents,heat,light,microwave,oxidant and common food additives have no effect on it,the metal ions Cu^2+,Mg^2+,Ca^2+,Zn^2+,Al2+and Fe^3+had no adverse effect on the stability of the melanin.The IR spectra showed that the melanin contained OH,n-h,C=O,C=C,carboxyl group,pyran ring,alcohol hydroxyl group and phenolic hydroxyl group.IR revealed that Cu^2+ion and Fe^3+ion chelated with the melanins were mainly complexated with the carboxyl and/or phenolic hydroxyl.These suggest S.reiliana has good stability and is a kind of natural pigment with great prospect of development and application,providing theoretical basis and methods for its further processing,utilization and development as a functional food.
