摘要
目的探讨冬凌草对非酒精性脂肪性肝病(NAFLD)大鼠血清生化指标和肝组织过氧化物酶体增殖剂激活受体(PPAR)α和PPARγ表达的影响。方法采用高脂饲料喂养的SD雄性大鼠构建NAFLD模型,随机分正常组、模型组、多烯磷脂酰胆碱组、冬凌草低、中和高剂量组,每组6只。HE染色观察大鼠肝组织脂肪变性程度;采用全自动生化仪测定生物化学指标;采用硫代巴比妥酸(TBA)法测定丙二醛(MDA)含量;采用黄嘌呤氧化酶法测定超氧化物歧化酶(SOD)活力;采用分光光度法测定谷胱甘肽过氧化物酶(GSH-Px)活力;提取肝组织采用免疫组织化学和RT-PCR测定各组大鼠肝组织PPARα、PPARγ蛋白和mRNA的表达。结果与模型组比较,冬凌草能降低ALT、AST活性和减少血清胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-H)的含量,升高高密度脂蛋白(HDL-C)的含量,调节血脂,显著增加肝组织SOD和GSH-Px活性,降低MDA含量(P均<0.05)。与正常组相比,模型组PPARα和PPARγ蛋白表达(6.95±4.45和2.73±1.07)及mRNA(0.36±0.02和0.21±0.02)明显减少(P<0.05),冬凌草低、中和高剂量组与模型组相比,PPARα、PPARγ蛋白及mRNA表达增多,并且呈剂量依赖性(P<0.05)。结论冬凌草对大鼠血清的生化指标和肝组织有良好的改善作用,这可能与其抗氧化、上调PPARα和PPARγ的表达有关。
Objective To investigate the effects of Rabdosia rubescens on the serum levels of biochemical markers and the expressions of peroxisome proliferator-activated receptors(PPAR)αandγin liver tissues of rats with non-alcoholic fatty liver disease(NAFLD).Methods The male SD rat models of NAFLD were established by high-fat diets.The rats were randomly divided into six groups,including normal group,model group,polyene phosphatidylcholine(Yishanfu)group,and Rabdosia rubescens low-dose,medium-dose and high-dose groups with 6 rats in each group.The degree of steatosis in liver tissues of rats was observed by HE staining.The biochemical parameters were measured by automatic biochemical analyzer.Malondialdehyde(MDA)was determined by thiobarbituric acid(TBA),superoxide dismutase(SOD)activity was determined by xanthione oxidase(XOD),and glutathione peroxidase(GSH-PX)activity was determined by spectrophotometry.The expressions of PPARα,PPARγprotein and mRNA in liver tissues of rats in each group were determined by immunohistochemistry and RT-PCR methods.Results Compared with the model group,Rabdosia rubescens reduced the activity of ALT and AST,decreased serum levels of total cholesterol(TC),total triglyceride(TG)and low-density lipoprotein cholesterol(LDL-C),and increased the level of high-density lipoprotein cholesterol(HDL-C)(P all<0.05).In addition,It regulated blood lipids,significantly increased the activity of SOD and GSH-Px of liver tissues,and reduced the MDA content(P all<0.05).Compared with the normal group,the expressions of PPARαand PPARγprotein(6.95±4.45 and 2.73±1.07)and mRNA(0.36±0.02 and 0.21±0.02)significantly decreased in the model group(P<0.05).Compared with the model group,the expressions of PPARαand PPARγprotein and mRNA increased in a dose-dependent manner in the Rabdosia rubescens low-dose,medium-dose and high-dose groups(P<0.05).Conclusions Rabdosia rubescens is effective in improving serum biochemical markers and liver tissues in rats,and it may be related to its anti-oxidation,up-regulatio
