一株具有抑菌活性的乳酸片球菌的鉴定与培养条件优化 被引量:3

Identification and optimization of culture conditions of abacteriostatic strain of Pediococcus acidilactici

摘要 为了开发一种高效南美白对虾微生态制剂,试验对前期从南美白对虾肠道中筛选到的1株抑菌性较好的菌株RSP-6进行菌落特征、亚显微形态、生理生化特征及16S rDNA同源序列分析,采用四因素(温度、pH值、氯化钠和碳源)三水平的正交试验设计,优化了该菌的培养条件。结果表明:RSP-6为乳酸片球菌。温度、pH值、氯化钠、碳源均显著影响乳酸片球菌的生长,且影响次序为pH值>氯化钠>碳源>温度。乳酸片球菌生长的最优条件为培养温度35℃,pH值6,氯化钠0,碳源为果糖。 In order to develop a highly effective microecological preparation for Penaeus vannamei, a strain RSP-6 with good bacteriostasis was screened from the intestinal tract of Penaeus vannamei. The strain RSP-6 was identified as Pediococcus acidilactici by the analysis of colony characteristics, submicroscopic morphology, physiological and biochemical characteristics and 16 SrDNA homology sequence analysis. With the absorbance value of P. acidilactici as the evaluation index, the culture conditions of P. acidilactici were optimized by orthogonal design of four factors(temperature, pH value, sodium chloride and carbon source) and three levels. The results showed that temperature, pH value, sodium chloride and carbon source all significantly affected the growth of P. acidilactici, and the order of influence was pH value>sodium chloride>carbon source>temperature. The optimum conditions for the growth of P. acidilactici were as follows: culture temperature 35 ℃, pH value 6, sodium chloride 0, and carbon source fructose.
