摘要
目的:通过网络药理学的手段研究西红花抗炎作用的分子机制。方法:搜集并整理得到西红花的有效成分信息,上传至Swiss数据库,匹配得到西红花有效成分对应的靶点蛋白,利用Cytoscape3.4.1软件构建有效成分-靶点的网络,对得到的靶点进行GO富集分析。通过TTD、CTD和OMIM数据库得到抗炎作用相关的靶点信息,建立疾病-靶点网络。选择Cytoscape 3.4.1中Merge功能对两个网络进行结合,得到有效成分-靶点-疾病网络。利用DAVID数据库对预测得到的靶点信息进行GO生物功能学富集分析和KEGG pathway富集分析。结果:筛选得到西红花抗炎活性成分14个,主要为类胡萝卜素类、黄酮类和酚酸类化合物。与抗炎作用相关的靶点有13个,其中度值较高的靶点有EGFR、KDR、PTPRC、APP、MET、MAPK12。推测西红花中活性成分通过EGFR、KDR、PTPRC、APP、MET、MAPK12等靶点参与了生物体内细胞信号传导、细胞代谢和基因表达等过程,抑制了炎症因子的产生和释放,从而发挥了抗炎作用。
Objective:To study the molecular mechanism of the anti-inflammatory effect of safflower by means of network pharmacology.Method:The active ingredient information of saffron were collected and organized and uploaded to the Swiss database,and then matched the target protein corresponding to the active ingredient of saffron.Cytoscape 3.4.1 software was used to construct an effective ingredient-target network,and then obtained the target Point for GO enrichment analysis.Through the TTD,CTD and OMIM databases,target information related to anti-inflammatory effects was obtained,and a disease-target network was established.Selected the Merge function in Cytoscape 3.4.1 to combine the two networks to get the effective component-target-disease network.The DAVID database was used to perform GO biofunctional enrichment analysis and KEGG pathway enrichment analysis on the predicted target information.Results:Screened 14 anti-inflammatory active ingredients of saffron,mainly carotenoids,flavonoids and phenolic compounds.There were 13 targets related to the anti-inflammatory effect,and the targets with higher degrees were EGFR,KDR,PTPRC,APP,MET,MAPK12.It is speculated that the active ingredients in safflower are involved in the process of cell signal transduction,cell metabolism and gene expression in the organism through EGFR,KDR,PTPRC,APP,MET,MAPK12 and other targets,inhibiting the production and release of inflammatory factors,thus exerting Anti-inflammatory effect.
