摘要
目的通过观察β-细辛醚对APP/PS1双转基因小鼠空间记忆能力及大脑海马神经突触相关功能蛋白突触后致密物质-95(PSD-95)表达的影响,探讨β-细辛醚对APP/PS1小鼠神经突触可塑性的保护作用及对阿尔茨海默病(AD)的治疗作用机制。方法选用APP/PS1双转基因小鼠40只,随机分为模型组、盐酸多奈哌齐组(0.33mg·kg^-1·d^-1)、β-细辛醚低剂量组(25mg·kg^-1·d^-1)及高剂量组(50mg·kg^-1·d^-1),选用相同月龄、相同遗传背景的C57BL/6小鼠10只为空白对照组。应用Morris水迷宫法检测小鼠的学习记忆能力,应用刚果红染色技术检测各组小鼠海马Aβ-淀粉酶沉积的变化,采用免疫荧光法检测小鼠海马区PSD-95蛋白的表达变化,RT-PCR技术检测各组小鼠海马区PSD-95mRNA表达水平。结果β-细辛醚能够明显改善APP/PS1双转基因小鼠的学习记忆功能障碍,提高学习与记忆功能,减少Aβ-淀粉酶沉积。免疫荧光及RT-PCR检测结果显示:β-细辛醚能够上调小鼠海马区PSD-95蛋白及m RNA的表达(P<0.05)。结论β-细辛醚具有改善AD的作用,并且作用机制与调控小鼠海马神经突触可塑性功能蛋白PSD-95的表达相关。
Objective To investigate the effect ofβ-asarone on the spatial memory ability of APP/PS1 double transgenic mice and the expression of post-synaptic dense substance-95(PSD-95)in hippocampal synapserelated functional protein,and study the protective effect ofβ-asarone on synaptic plasticity in APP/PS1 mice and therapeutic mechanism of action on Alzheimer’s disease(AD).Methods Forty APP/PS1 double transgenic mice were randomly divided into model group,donepezil hydrochloride group(0.33 mg·kg^-1·d^-1),β-asarone low dose group(25 mg·kg^-1·d^-1)and high dose group(50 mg·kg^-1·d^-1),10 C57 BL/6 mice of the same age and the same genetic background were selected as blank control group.The learning and memory ability of mice were detected by Morris water maze method.The changes of Aβ-amylase deposition in hippocampus of each group were observed by Congo red staining.The expression of PSD-95 protein in hippocampus of mice was detected by immunofluorescence.And RT-PCR technology was used to detect the expression of PSD-95 mRNA in hippocampus of each group.Results β-asarone could significantly improve learning and memory dysfunction of APP/PS1 double transgenic mice,improve learning and memory functions,and reduce Aβ-amylase deposition.Immunofluorescence and RT-PCR showed thatβ-asarone could increase the protein and mRNA expression of PSD-95 in hippocampus of mice(P<0.05).Conclusionβ-asarone has the effect of improving AD,and the mechanism of action is related to the regulation of the expression of functional protein PSD-95 of hippocampal synaptic plasticity in mice.
