响应面法优化辣木籽蛋白质提取工艺的研究 被引量:5

摘要 以脱脂辣木籽粉为原料,采用超声波辅助碱法提取辣木籽蛋白质,运用单因素和响应面法优化辣木籽蛋白质提取工艺参数。结果表明,其最佳提取工艺条件为:pH 8.0、提取时间45 min、提取温度44℃、超声功率150 W,按照此工艺参数,辣木籽蛋白质提取率达44.03%。各因素影响辣木籽蛋白质提取率顺序为:超声功率>提取时间>提取温度>pH值。辣木籽蛋白质等电点为3.4,沉淀后辣木籽蛋白质沉淀率和纯度分别为54.27%、91.31%。辣木籽蛋白质SDS-PAGE分析表明,其主要含有分子量为16.0、25.0和45.0kDa的3个亚基。氨基酸分析表明,该蛋白质中含有18种氨基酸,其中7种是人体必需氨基酸,占总氨基酸含量的25.63%,谷氨酸含量最高,达到9.031g/(100g)。 With degreased moringa oleifera seed powder as raw material,the protein of Moringa oleifera seed was extracted by ultrasonic assisted alkali method,and the technological parameters of protein extraction of Moringa oleifera seed were optimized by single factor and response surface method.The results showed that the optimal extraction conditions were as follows:pH 8.0,extraction time 45 min,extraction temperature 44℃,ultrasonic power 150 W.According to the process parameters,the protein extraction rate of moringa oleifera seed reached 44.03%.The sequence of factors influencing the protein extraction rate of Moringa oleifera seed was as follows:ultrasonic power>extraction time>extraction temperature>pH value.The isoelectric point of Moringa oleifera seed protein was 3.4,the precipitation rate was 54.27%and purity of Moringa oleifera seed protein was 91.31%after precipitation.The SDS PAGE analysis of Moringa oleifera seed protein showed that it mainly contained three subunits with molecular weights of 16.0,25.0 and 45.0 kDa.Amino acid analysis indicated that Moringa oleifera seed protein contained 18 kinds of amino acids,of which 7 kinds were the essential amino acids,accounting for 25.63%of the total amino acid content,and glutamic acid content was the highest,reaching 9.031 g/(100 g).
