基于网络药理学探讨交泰丸治疗失眠的作用机制 被引量:17

摘要 目的运用网络药理学研究方法探讨交泰丸治疗失眠症的作用机制。方法应用中药药理学平台TCMSP数据库和Batman数据库筛选交泰丸的成分和相关靶点,使用Uniprot数据库,将靶点名称进行矫正统一,并使用Swiss Target Prediction数据库对靶点信息进行补充;并通过Gene Cards数据库以“失眠”为关键词进行检索,获得疾病作用靶点;在Venny 2.1在线软件作图工具平台上分别录入黄连、肉桂与失眠症的靶点,绘制韦恩图,得到药物与疾病的共同靶点;使用Cytoscape 3.7.1软件,构建“药物-成分-疾病-靶点”相互作用网络图,使用Network Analyzer功能对中药复方的主要活性成分进行分析;将共同靶点输入到STRING数据库中进行检索,构建蛋白相互作用关系(PPI)网络,并根据蛋白之间的关联度进行排序;在R软件安装Bioconductor软件包“org.Hs.eg.db”,将药物-疾病共同靶点转换成entrezID,然后在R软件安装“clusterProfiler”包,根据已转换的entrezID,以P<0.05,Q<0.05进行关键靶基因的基因本体论(GO)与京都基因和基因组百科全书(KEGG)功能富集分析。结果筛选出交泰丸34个有效成分和498个药物靶点,失眠疾病靶点2279个;通过韦恩图取交集后获得药物-疾病共同靶点160个,筛选出黄连-肉桂与失眠症共同靶点基因21个;这些共同靶点涉及第二信使介导的信号传递,G蛋白偶联受体信号传导途径、与环状核苷酸第二信使偶联,对细菌起源分子的反应,调节膜电位,调节炎症反应等10余个重要的生物学过程;获得生物学通路包括Toll样受体信号通路、核因子-κB(NF-κB)信号通路、环磷酸腺苷(cAMP)信号通路、γ-氨基丁酸(GABA)能突触、肿瘤坏死因子(TNF)信号通路、胰岛素抵抗、MAPK信号通路、cGMP-PKG信号通路等重要通路。结论交泰丸可能通过调节炎症通路、干预炎症的发生发展以及改善胰岛素抵抗来发挥治疗失眠的作用。 Objective To explore the mechanism of Jiaotai Pill in treating insomnia based on network pharmacology research methods.Methods The TCMSP database and Batman database were used to screen the components of Jiaotai Pills and related targets.The Uniprot database was used to correct the target names.The Swiss Target Prediction database was used to supplement the target information."Insomnia"was the keyword,the disease targets were searched by Gene Cards database.The target of coptis chinensis,cinnamon,and insomnia were input respectively on the Venny2.1 online software mapping tool platform,the Wayne diagram were drawn,and the common target of drugs and diseases were got.The"drug-component-disease-target"interaction network diagram were constructed by Cytoscape 3.7.1 software.The main active ingredients of the traditional Chinese medicine compound were analyzed by the Network Analyzer function.The common targets were entered into the STRING database for retrieval,and the PPI network of protein interactions were constructed.Bioconductor software package"org.Hs.eg.db"in R software was install,the common drug-disease target was converted into entrezID,and then the"clusterProfiler"package was installed in R software.The perform key targets were enriched by GO and KEGG functional analysis with P<0.05 and Q<0.05 based on the converted entrezID.Results Thirty-four active ingredients and 498 drug targets were selected for Jiaotai Pills,and 2279 disease targets for insomnia were screened out.One hundred and sixty drug-disease common targets were obtained after the intersection of the Venn diagrams,and 21 Coptis-cinnamon and insomnia common target genes were screened.These common targets involved more than 10 important biological processes such as second-messenger-mediated signaling,G protein-coupled receptor signaling pathway,coupled to cyclic nucleotide second messenger,response to molecule of bacterial origin,regulation of membrane potential,regulation of inflammatory response and so on.A total of 8 important signaling pathw
