摘要
目的研究EZH2对鼻咽癌血管生成拟态的影响及可能机制.方法慢病毒载体pLVTHM建立空载体对照(Lv-vector)、稳定过表达EZH2(Lv-EZH2)和沉默EZH2(Lv-shEZH2)的鼻咽癌细胞株;检测EZH2对鼻咽癌细胞增殖、迁移和血管生成拟态的影响;检测EZH2调控的下游相关的靶基因的表达.结果相比空载体组(Lv-vector),Lv-EZH2组细胞生长速度、相对移动距离、穿过聚碳酸酯膜的细胞数、单位面积内成管数、靶基因(ET-1、NOTCH4、CXCL5、HPSE)表达显著增加,而靶基因(PLAU、KDR、THBS1)表达显著降低(P<0.05);Lv-shEZH2组细胞生长速度、相对移动距离、穿膜的细胞数、单位[面积内成管数、靶基因(ET-1、NOTCH4、CXCL5、HPSE)表达显著降低,而靶基因(PLAU、KDR、THBS1)表达显著增加(P<0.05).结论EZH2可促进鼻咽癌细胞体外的增殖、迁移和血管生成拟态,机制与多个下游基因调控有关.
Objective To explore the effect of EZH2 on vasculogenic mimicry of nasopharyngeal carcinoma(NPC)and its possible mechanism.Methods Lentiviral vector(pLVTHM)was used to establish NPC cell lines with empty vector control(LV-vector),stably overexpressed EZH2(LV-EZH2)and silencing EZH2(LV-shEZH2).The effects of EZH2 on the proliferation,migration and vasculogenic mimicry of NPC cells were detected and the expression of downstream target genes regulated by EZH2 was detected.Results Compared with LV-vector group,LV-EZH2 group showed significantly increased cell growth rate,relative moving distance,number of cells passing through polycarbonate membrane,number of tube formation per unit area and the expression of target genes ET-1,NOTCH4,CXCL5 and HPSE,and significantly decreased expression of target genes PLAU,KDR and THBS1(P<0.05);LV-shEZH2 group showed significantly decreased cell growth rate,relative moving distance,number of cells passing through polycarbonate membrane,number of tube formation per unit area and the expression of target genes ET-1,NOTCH4,CXCL5 and HPSE,and significantly increased expression of target genes PLAU,KDR and THBS1(P<0.05).Conclusion EZH2 can promote the proliferation,migration and vasculogenic mimicry of NPC cells in vitro,and its mechanism is correlated with the regulation of several downstream genes.
