摘要
目的观察柴胡三参胶囊对缺血性心律失常大鼠心肌细胞Bcl-2及Fas蛋白的表达影响,进一步明确柴胡三参胶囊治疗缺血性心律失常的作用及机制。方法将实验大鼠随机分为假手术组、模型组、柴胡三参胶囊组、参松养心胶囊组、维拉帕米组、空白组,每组各10只。给予相应药物干预2周后观察各组的心律失常积分和Bcl-2、Fas表达的变化。结果柴胡三参胶囊组、参松养心胶囊组、维拉帕米组与模型组进行比较,心律失常评分下降(P <0.05),Bcl-2蛋白表达升高(P <0.01),Fas蛋白表达有一定程度降低(P <0.05)。结论柴胡三参胶囊能上调抑制凋亡因子Bcl-2表达,下调促进凋亡因子Fas表达,说明其可减轻细胞凋亡,缓解心肌损伤,为柴胡三参胶囊改善缺血性心律失常提供了实验依据。
Objective:To observe the effect of Chaihu Sanshen Capsule on the expression of Bcl-2 and Fas protein in myocardial cells of rats with ischemic arrhythmia and to clarify the effect and mechanism of Bupleurum Sanshen capsule in the treatment of ischemic arrhythmia.Methods:The experimental rats were randomly divided into sham operation group,model group,Chaihu Sanshen Capsule group,Shensong Yangxin Capsule group,verapamil group,blank group,10 in each group.The changes of arrhythmia scores and Bcl-2,fas expression in the six groups were observed.Results:Compared with the model group,the arrhythmia scores of Chaihu Sanshen Capsule group,Shensong Yangxin Capsule group and verapamil group decreased significantly(P<0.05).The expression of Bcl-2 protein was increased,the drug intervention was effective,and the difference was statistically significant(P<0.01).Compared with the model group,the expression of Fas protein was reduced to a certain extent,and the difference was statistically significant(P<0.05).Conclusion:Chaihu Sanshen Capsule up-regulates and inhibits the expression of apoptosis factor Bcl-2,down-regulates and promotes the expression of apoptosis factor Fas,indicating that it can alleviate apoptosis and relieve myocardial injury,which provides experimental basis for Chaihu Sanshen Capsule to improve ischemic arrhythmia.
