地五养肝方基于肝肾同源对肝纤维化大鼠肝功能及肝纤维化的影响研究 被引量:5

Effect and Mechanism of Diwu Yanggan Recipe on Liver Function and Liver Fibrosis for Rats with Hepatic Fibrosis

摘要 目的:分析肝肾同源理论下地五养肝方对肝纤维化大鼠肝功能及肝纤维化影响。方法:选取SPF级Wistar雄性大鼠60只,随机数字表法将大鼠分成4组,分别为正常组、模型组、阳性对照组和地五养肝方组,除正常组外,其他各组大鼠制备肝纤维化模型,大鼠首次注射的第2 d起,阳性对照组灌胃0.11mg/kg的秋水仙碱混悬液,地五养肝方组灌胃2.2g/kg的地五养肝方药液,模型组和正常组灌胃等剂量生理盐水,1次/d,连续灌胃8周。观察大鼠肝组织病理形态和肝纤维化程度,ELISA法检测血清肝功能指标和相关细胞因子含量,Western-blot检测大鼠肝组织p-AKT、β-catenin、AKT、p-GSK-3β、GSK-3β蛋白表达。结果:模型组大鼠肝纤维化分级较正常组显著上升,阳性对照组、地五养肝方组大鼠肝纤维化分级较模型组显著下降,差异有统计学意义(P<0.05);和正常组相比,模型组大鼠血清GLB、ALB、TP含量降低,ALT、AST含量升高;和模型组相比,阳性对照组、地五养肝方组大鼠血清GLB、ALB、TP含量升高,ALT、AST含量降低,差异有统计学意义(P<0.05);和正常组相比,模型组大鼠肝组织β-catenin、p-GSK-3β及p-AKT蛋白表达升高;和模型组相比,阳性对照组、地五养肝方组大鼠肝组织β-catenin、p-GSK-3β及p-AKT蛋白表达降低,差异有统计学意义(P<0.05)。结论:地五养肝方可明显降低大鼠肝纤维程度,其作用机制可能和对Wnt/β-catenin信号通路的活化抑制有联系。 Objective:To analyze the effect of Diwu Yanggan Recipe on liver function and liver fibrosis for rats with hepatic fibrosis and its related mechanisms.Methods:Sixty Wistar male rats of SPF grade were randomly divided into four groups:normal group,model group,positive controlled group and Diwu Yanggan group.Except the normal group,the other groups with rat model of liver fibrosis was prepared.On the 2 nd day after the first injection of the rats,the positive controlled group was intragastrically administered with 0.11 mg/kg colchicine suspension,and the Diwu Yanggan Recipe group was administered with 2.2 g/kg of the ground.Liver medicine,model group and normal group were intragastrically administered with normal saline once a day for 8 weeks.The pathological morphology and degree of liver fibrosis of rat liver were observed.Serum liver function index and related cytokines were detected by ELISA.Western blotting was used to detect p-AKT,β-catenin,AKT and p-GSK-3βin rat liver tissue GSK-3βprotein expression.Results:The liver fibrosis grade of the model group was significantly higher than that of the normal group.The liver fibrosis grade of the rats in the positive controlled group and the Diwu Yanggan Recipe group was significantly lower than that in the model group(P<0.05).Compared with the normal group,the serum levels of GLB,ALB and TP in the model group decreased,and the levels of ALT and AST increased.Compared with the model group,the serum levels of GLB,ALB and TP in the positive controlled group and Diwu Yanggan Recipe group were higher.The levels of ALT and AST decreased(P<0.05).Compared with the normal group,the expression ofβ-catenin,p-GSK-3βand p-AKT protein in the model group increased.Compared with the model group,the expressions ofβ-catenin,p-GSK-3βand p-AKT protein in the liver of rats in the positive controlled group and Diwu Yanggan Recipe group were significantly lower(P<0.05).Conclusion:Diwu Yanggan Recipe can significantly reduce the degree of liver fibrosis in rats,and its mechanism may be
